[目的]利用巴马小香猪构建空肠弯曲菌致GBS的动物模型。[方法]选取致人GBS源空肠弯曲菌株,采用3×1012 cfu/mL、3×1011 cfu/mL、3×1010 cfu/mL、3×109 cfu/mL等浓度对4组巴马小型猪灌胃攻毒,每头猪攻毒10mL,同时设立对照组。从第二周开始定期采集组织样及血液样本,通过免疫学检测及组织病理切片判定病变情况。[结果]免疫学检测发现,在16份攻毒组样品中有12份血清中GM1-IgG抗体检测阳性,阳性检出率为81.25%。但是,并没有显著的攻毒浓度梯度差异。病理组织切片H/E染色显示,抗体阳性试验猪的大脑出现了噬神经元、小血管周围间隙增宽、神经中央染色溶解等现象;切片LFB染色显示,在发病模型动物的小脑白质和腰膨大出现了轻度脱髓鞘现象。[结论]结果表明空肠弯曲菌诱导GBS的浓度在3×109 cfu/mL~3×1012 cfu/mL均可,组织样本及血液采集在攻毒后4~5周较理想。
[目的]利用巴馬小香豬構建空腸彎麯菌緻GBS的動物模型。[方法]選取緻人GBS源空腸彎麯菌株,採用3×1012 cfu/mL、3×1011 cfu/mL、3×1010 cfu/mL、3×109 cfu/mL等濃度對4組巴馬小型豬灌胃攻毒,每頭豬攻毒10mL,同時設立對照組。從第二週開始定期採集組織樣及血液樣本,通過免疫學檢測及組織病理切片判定病變情況。[結果]免疫學檢測髮現,在16份攻毒組樣品中有12份血清中GM1-IgG抗體檢測暘性,暘性檢齣率為81.25%。但是,併沒有顯著的攻毒濃度梯度差異。病理組織切片H/E染色顯示,抗體暘性試驗豬的大腦齣現瞭噬神經元、小血管週圍間隙增寬、神經中央染色溶解等現象;切片LFB染色顯示,在髮病模型動物的小腦白質和腰膨大齣現瞭輕度脫髓鞘現象。[結論]結果錶明空腸彎麯菌誘導GBS的濃度在3×109 cfu/mL~3×1012 cfu/mL均可,組織樣本及血液採集在攻毒後4~5週較理想。
[목적]이용파마소향저구건공장만곡균치GBS적동물모형。[방법]선취치인GBS원공장만곡균주,채용3×1012 cfu/mL、3×1011 cfu/mL、3×1010 cfu/mL、3×109 cfu/mL등농도대4조파마소형저관위공독,매두저공독10mL,동시설립대조조。종제이주개시정기채집조직양급혈액양본,통과면역학검측급조직병리절편판정병변정황。[결과]면역학검측발현,재16빈공독조양품중유12빈혈청중GM1-IgG항체검측양성,양성검출솔위81.25%。단시,병몰유현저적공독농도제도차이。병리조직절편H/E염색현시,항체양성시험저적대뇌출현료서신경원、소혈관주위간극증관、신경중앙염색용해등현상;절편LFB염색현시,재발병모형동물적소뇌백질화요팽대출현료경도탈수초현상。[결론]결과표명공장만곡균유도GBS적농도재3×109 cfu/mL~3×1012 cfu/mL균가,조직양본급혈액채집재공독후4~5주교이상。
Objective] Bama mini-pig was used to establish the animal model for C. jejuni induced GBS. [Methods] C. jejuni isolate inducing GBS in human was selected to infect pigs. Four groups,each with four pigs,were orally infused respectively with four concentration bacterial suspension of 3×1012 cfu/mL,3×1011 cfu/mL,3×1010 cfu/mL and 3×109 cfu/mL. A control group was set up and orally infused with normal saline. From the second week,blood and tissue samples were collected regularly to determine the immune reaction and lesions by ELISA and biopsy. [Results] 12 of 16 samples in the infected groups were positive for GM1-IgG antibodies with a positive rate of 81.25%. However, there was no significant difference between the groups infected with different concentration gradients. The H/E staining revealed neuronophagia,enlarged Virchow-Robin spaces,central chromatolysis and so on in the cerebrum of the infected animals;LFB staining showed mildly demyeliantion in the cerebellar white matter and the lumbar enlargement of the infected animals. [Conclusion]The result showed that the C. jejuni concentration for inducing GBS in mini-pigs was 3×109 cfu/mL~3×1012 cfu/mL and the best time to collect blood and tissue samples was 4~5 weeks after inoculation.
