分别采用木瓜蛋白酶和溶壁酶降解金针菇菇根水提后的多糖,以提取还原性寡糖,并通过单因素试验和正交试验确定适合两种酶的最佳提取工艺。结果表明,木瓜蛋白酶降解菇根多糖的最佳工艺条件为酶解0.5 h、酶底比为4.5%、pH值6.0、温度为35℃;溶壁酶的最佳工艺条件为酶解1.5 h、酶底比为2.0%、pH值5.0、温度为50℃。采用不同的酶降解金针菇多糖,其还原性寡糖的得率不同,其中,溶壁酶降解所得还原性寡糖得率为6.8768%,木瓜蛋白酶降解所得还原性寡糖得率为6.1368%。

분별채용목과단백매화용벽매강해금침고고근수제후적다당,이제취환원성과당,병통과단인소시험화정교시험학정괄합량충매적최가제취공예。결과표명,목과단백매강해고근다당적최가공예조건위매해0.5 h、매저비위4.5%、pH치6.0、온도위35℃;용벽매적최가공예조건위매해1.5 h、매저비위2.0%、pH치5.0、온도위50℃。채용불동적매강해금침고다당,기환원성과당적득솔불동,기중,용벽매강해소득환원성과당득솔위6.8768%,목과단백매강해소득환원성과당득솔위6.1368%。
In order to obtain oligosaccharides , the papain and lywallzyme were used respectively to de-grade the polysaccharides extracted from enoki mushroom root with water .The optimum enzymolysis conditions for two enzymes were optimized through single -factor experiment and orthogonal test .The results showed that the optimum conditions for papain were enzymolysis for 0.5 hours, the ratio of enzyme to substrate as 4.5%, pH 6.0 and the temperature as 35℃.The optimum conditions for lywallzyme were enzymolysis for 1.5 hours, the ratio of enzyme to substrate as 2.0%, pH 5.0 and the temperature as 50℃.Using different enzymes to degrade enoki mushroom polysaccharides , the production of oligosaccharides was different .The yield of reduc-ing oligosaccharide obtained by lywallzyme and papain were 6.8768%and 6.1368%respectively.
