目的:探讨维生素D受体在内毒素感染时对肝脏的保护作用。方法随机抽取C57 BL/6源性的野生小鼠10只和维生素D受体( VDR)敲除小鼠10只,分为对照组和实验组,实验组LPS 15 mg/kg腹腔注射。应用免疫组化及免疫荧光法、实时定量PCR法等方法测定小鼠血浆中的谷草转氨酶含量、细胞因子及进行PAI-1在肝脏组织中的定量分析。结果注射LPS后,VDR敲除鼠肝脏中的巨噬细胞阳性染色明显多于对照组。 VDR敲除组血浆AST水平明显高于对照组。在基因敲除鼠肝脏中伤害性细胞因子IL-2、IL-18、IL-12、IFN-γ明显升高,而保护性细胞因子IL-10明显降低。 PAI-1在VDR敲除鼠的肝脏中表达明显增多。结论 VDR在内毒素对肝脏损伤时能够起到保护作用,为进一步研究维生素D及其信号系统的功能提供了理论依据。

목적:탐토유생소D수체재내독소감염시대간장적보호작용。방법수궤추취C57 BL/6원성적야생소서10지화유생소D수체( VDR)고제소서10지,분위대조조화실험조,실험조LPS 15 mg/kg복강주사。응용면역조화급면역형광법、실시정량PCR법등방법측정소서혈장중적곡초전안매함량、세포인자급진행PAI-1재간장조직중적정량분석。결과주사LPS후,VDR고제서간장중적거서세포양성염색명현다우대조조。 VDR고제조혈장AST수평명현고우대조조。재기인고제서간장중상해성세포인자IL-2、IL-18、IL-12、IFN-γ명현승고,이보호성세포인자IL-10명현강저。 PAI-1재VDR고제서적간장중표체명현증다。결론 VDR재내독소대간장손상시능구기도보호작용,위진일보연구유생소D급기신호계통적공능제공료이론의거。
Objective To explore the protective effect of Vitamin D receptor on hepatic injury induced by LPS. Methods The C57 BL/6 mice were randomly divided into control group and experimental group from 10 wild and 10 Vitamin D receptor ( VDR) knockout mice respectively. LPS 15 mg/kg i. p. was injected into experimental group. Plasma AST of mice,the change of cytokines and PAI-1 quantity in liver tissue were determined by immunofluo-rescence, real-time PCR methods. Results Marked accumulation of macrophages were more in the liver of VDR knockout mice after LPS-treated than those in control group. Plasma AST in VDR knockout mice was significantly higher than that in control group. Nocuous cytokines IL-2,IL-18,IL-12,IFN-γ were significantly higher,and protective cytokine IL-10 was significantly lower in the liver of VDR knockout mice than those in control group. PAI-1 was sig-nificantly higher in the liver of VDR knockout mice. Conclusion Vitamin D receptor can play a protective role on he-patic injury induced by LPS,which provides a theoretical evidence for further study of Vitamin D and the function of its signal function.
