目的:通过观察CPC/BMP复合材料在动物实验中对眼球形态的影响,并检测其体外细胞毒性,评价其对眼的毒性。方法将CPC/BMP复合材料及对照组单纯CPC材料分别植入18只新西兰兔右、左眼眶骨缺损区,每日观察动物眼外观有无异常。使用植入眼眶后0天、3天和3周的CPC/BMP复合材料、单纯CPC材料的浸提液、Medpor材料浸提液、单纯培养液、0.64%苯酚稀释液培养L929小鼠肺成纤维细胞和人角膜缘上皮细胞,孵育1天、3天时,MTT法检测吸光度(OD值),并计算细胞相对增值率,判定细胞毒性。结果 CPC/BMP复合材料及单纯CPC材料植入眼眶骨缺损区术后可见实验动物出现一过性结膜充血。各时间点CPC/BMP复合材料、单纯CPC材料、Medpor材料及空白对照组的OD值差异无统计学意义(P>0.05),细胞相对增值率≥80%,细胞毒性≤1级。结论 CPC/BMP复合材料未见明显眼毒性,符合眼眶内植入材料的生物学要求。

목적:통과관찰CPC/BMP복합재료재동물실험중대안구형태적영향,병검측기체외세포독성,평개기대안적독성。방법장CPC/BMP복합재료급대조조단순CPC재료분별식입18지신서란토우、좌안광골결손구,매일관찰동물안외관유무이상。사용식입안광후0천、3천화3주적CPC/BMP복합재료、단순CPC재료적침제액、Medpor재료침제액、단순배양액、0.64%분분희석액배양L929소서폐성섬유세포화인각막연상피세포,부육1천、3천시,MTT법검측흡광도(OD치),병계산세포상대증치솔,판정세포독성。결과 CPC/BMP복합재료급단순CPC재료식입안광골결손구술후가견실험동물출현일과성결막충혈。각시간점CPC/BMP복합재료、단순CPC재료、Medpor재료급공백대조조적OD치차이무통계학의의(P>0.05),세포상대증치솔≥80%,세포독성≤1급。결론 CPC/BMP복합재료미견명현안독성,부합안광내식입재료적생물학요구。
Objective To determine the ocular toxicity of the composite of CPC loaded with BMP. Methods CPC-BMP composites and CPC control were implanted in orbital fracture of 18 rabbits, and then the eyeball were evaluated everyday. In the cytotoxicity assays, mouse pulmonary fibroblasts( L929) and human corneal epithelial cell (HCEC) were incubated in the extracts of CPC-BMP composites,CPC control and Medpor control which were implanted in the orbital fracture 0 day, 3 days and 3 weeks. Then at the end of the extract incubation period(1 day and 3 day) ,the cellular function was assessed with MTT assay.Results Both CPC-BMP composites and CPC control could induce the conjunctival congestion in a short period.CPC-BMP composites had no apparent cytotoxicity compared with CPC ontrol , medpor control,blank control and negative control with or without implanted in the orbital. Conclusion The composites of CPC loaded with BMP has no apparent ocular toxicity. It is suitable for use in the orbital, the same as Medpor.
