目的:研究姜黄素体外对人结肠癌Lovo细胞血管内皮生长因子VEGF表达的影响。方法不同浓度(0μmol/L、5μmol/L、10μmol/L、20μmol/L、40μmol/L)姜黄素处理2013年8月—2014年2月中南大学提供的人结肠癌细胞株Lovo24 h后,用Real-time PCR检测姜黄素对人结肠癌细胞株血管内皮生长因子VEGF mRNA表达的变化,用Western blot法检测姜黄素对人结肠癌细胞株VEGF蛋白表达的变化。结果人结肠癌Lovo细胞经过姜黄素处理24 h后,5μmol/L、10μmol/L、20μmol/L、40μmol/L姜黄素组VEGF mRNA及蛋白的表达均明显下降,与对照组比较差异有统计学意义(P<0.05)。结论姜黄素能下调人结肠癌Lovo细胞VEGF的表达,并呈剂量依赖,这可能是姜黄素抑制结肠癌Lovo细胞增殖及侵袭性的机制之一。

Objective To study the effect of curcumin on the expression of VEGF in human colon cancer Lovo cells in vitro. Methods The human colon cancer cell lines Lovo were treated with 0 μmol/L,5 μmol/L10 μmol/L,20 μmol/L, 40 μmol/L curcum-in for 24 hours respectively, then we detected the human colon cancer cell lines VEGF mRNA expression by Real-time PCR and VEGF protein expression by Western Blot. Results After the human colon cancer cell lines Lovo cells treated with curcumin for 24 hours respectively, the expression of VEGF mRNA and protein of the curcumin-treated groups (5 μmol/L,10 μmol/L,20 μmol/L, 40 μmol/L) had more significant reduction compared with control group (P<0.05). Conclusion Curcumin could decrease the ex-pression of VEGF in human colon cancer Lovo cells dose-dependently, this may provide some evidence for the mechanism that curcumin could inhibit the proliferation and invasion of colon cancer Lovo cells.
