目的:观察抑郁症大鼠模型脑组织不同部位神经营养因子水平的变化及抗抑郁药物使用后改变。方法:将成年SD大鼠30只分为正常对照组、模型对照组、药物干预组,每组10只,雌雄对半;正常组正常养殖,模型组采用慢性不可预知应激结合孤养方式制备抑郁模型大鼠4周。药物干预组采用模型组造模过程4周后给予药物(氟西汀)给入,自实验开始之后每周观察大鼠体重、糖水消耗、旷场实验指标的变化,最后采用荧光定量PCR法观察大鼠脑部BDNF因子及Trkb的表达。结果:与正常对照组大鼠相比,模型组大鼠体重增加缓慢、糖水消耗减少、旷场试验各项指标较正常组间差异具有统计学意义;荧光定量PCR结果显示:模型组较对照组各脑区BDNF及Trkb受体表达减少,药物干预组脑区BDNF及Trkb受体表达显著高于模型对照组。结论:脑部BDNF因子及Trkb的表达在抑郁症发生与治疗中有所改变。
目的:觀察抑鬱癥大鼠模型腦組織不同部位神經營養因子水平的變化及抗抑鬱藥物使用後改變。方法:將成年SD大鼠30隻分為正常對照組、模型對照組、藥物榦預組,每組10隻,雌雄對半;正常組正常養殖,模型組採用慢性不可預知應激結閤孤養方式製備抑鬱模型大鼠4週。藥物榦預組採用模型組造模過程4週後給予藥物(氟西汀)給入,自實驗開始之後每週觀察大鼠體重、糖水消耗、曠場實驗指標的變化,最後採用熒光定量PCR法觀察大鼠腦部BDNF因子及Trkb的錶達。結果:與正常對照組大鼠相比,模型組大鼠體重增加緩慢、糖水消耗減少、曠場試驗各項指標較正常組間差異具有統計學意義;熒光定量PCR結果顯示:模型組較對照組各腦區BDNF及Trkb受體錶達減少,藥物榦預組腦區BDNF及Trkb受體錶達顯著高于模型對照組。結論:腦部BDNF因子及Trkb的錶達在抑鬱癥髮生與治療中有所改變。
목적:관찰억욱증대서모형뇌조직불동부위신경영양인자수평적변화급항억욱약물사용후개변。방법:장성년SD대서30지분위정상대조조、모형대조조、약물간예조,매조10지,자웅대반;정상조정상양식,모형조채용만성불가예지응격결합고양방식제비억욱모형대서4주。약물간예조채용모형조조모과정4주후급여약물(불서정)급입,자실험개시지후매주관찰대서체중、당수소모、광장실험지표적변화,최후채용형광정량PCR법관찰대서뇌부BDNF인자급Trkb적표체。결과:여정상대조조대서상비,모형조대서체중증가완만、당수소모감소、광장시험각항지표교정상조간차이구유통계학의의;형광정량PCR결과현시:모형조교대조조각뇌구BDNF급Trkb수체표체감소,약물간예조뇌구BDNF급Trkb수체표체현저고우모형대조조。결론:뇌부BDNF인자급Trkb적표체재억욱증발생여치료중유소개변。
Objective: To observe the depression rat model of brain tissue in different parts of the neurotrophic factor levels change and the use of antidepressants after the change.Methods:30 adult SD rats were divided into normal control group, model group, drug intervention groups of 10 male and female in half; normal normal breeding, the model group with chronic unpredictable stress dependent manner combined with solitary depression in rats 4 weeks. Drug intervention group with model group modeling process four weeks after administration of the drug (fluoxetine) to enter, since the beginning of the experiment rats were observed weekly body weight, sugar consumption,changes in open-field test indicators, last observed by fluorescence quantitative PCR rat brain BDNF expression of cytokines and Trkb.Results:Compared with normal control group, model group rats slow weight gain, sugar consumption is reduced, between open-field test of the indicators compared with the normal group, a statistically significant difference; quantitative PCR results showed that: the model group than in the control group various brain regions decreased expression of BDNF and Trkb receptors, drug intervention group BDNF and its receptor expression in brain regions Trkb significantly higher than the model group.Conclusion:The expression of BDNF factor and Trkb brain occurs in depression and treatment change.
