目的:观察胰岛素样生长因子-1(IGF-1)及其受体(IGFR-1)信号通路在前列腺癌中的作用。方法取50例前列腺癌患者石蜡包埋的前列腺癌组织(前列腺癌组),标本经行根治性前列腺切除术留取;并与50例良性前列腺增生组织(良性前列腺增生组)对照,标本经行尿道前列腺电切除术留取。免疫组化法测定两组前列腺组织IGF-1、IGFR-1蛋白的表达。结果前列腺癌组IGF-1、IGFR-1蛋白的阳性表达率均显著高于良性前列腺增生组(依次为:56%比30%、60%比38%,P<0.05)。前列腺癌组IGF-1、IGFR-1蛋白的阳性表达强度(依次为:0.168±0.016、0.174±0.022OD值)也均显著高于良性前列腺增生组(依次为:0.135±0.024、0.150±0.027OD值)(P<0.01)。相关分析显示,IGF-1/IGFR-1蛋白的阳性表达强度无显著相关性(r=0.242,P>0.05)。结论前列腺癌组织中IGF-1/IGFR-1的表达水平增强,IGF/IGFR信号系统可作为前列腺癌预后的预测指标。

목적:관찰이도소양생장인자-1(IGF-1)급기수체(IGFR-1)신호통로재전렬선암중적작용。방법취50례전렬선암환자석사포매적전렬선암조직(전렬선암조),표본경행근치성전렬선절제술류취;병여50례량성전렬선증생조직(량성전렬선증생조)대조,표본경행뇨도전렬선전절제술류취。면역조화법측정량조전렬선조직IGF-1、IGFR-1단백적표체。결과전렬선암조IGF-1、IGFR-1단백적양성표체솔균현저고우량성전렬선증생조(의차위:56%비30%、60%비38%,P<0.05)。전렬선암조IGF-1、IGFR-1단백적양성표체강도(의차위:0.168±0.016、0.174±0.022OD치)야균현저고우량성전렬선증생조(의차위:0.135±0.024、0.150±0.027OD치)(P<0.01)。상관분석현시,IGF-1/IGFR-1단백적양성표체강도무현저상관성(r=0.242,P>0.05)。결론전렬선암조직중IGF-1/IGFR-1적표체수평증강,IGF/IGFR신호계통가작위전렬선암예후적예측지표。
Objective To investigate the potential roles of insulin-like growth factor-I (IGF-I) and it's type I receptor (IGFR-I) in the prostate neoplastic tissues. Methods Fifty paraffin-embedded prostatic cancer samples (prostatic cancer group) which were obtained by radical prostatectomy were analyzed by using immunohistochem-istry. Other 50 samples of benign prostatic hyperplasia (benign prostatic hyperplasia group) which were obtained by transurethral resection of prostate served as control. Results The positive expression rate of IGF-I and IGFR-I in prostatic cancer group were significantly higher than those in benign prostatic hyperplasia group(56% vs 30%, 60%vs 38%, respectively; P all <0.05). The expression levels of them in prostatic cancer group (0.168±0.016; 0.174± 0.022 optical density,respectively) were dramatically higher than those in benign prostatic hyperplasia group (0.135 ±0.024; 0.150 ±0.027 optical density, respectively; all P<0.01). There was no statistic correlation between IGF-1 and IGFR-1 expression in two groups(r=0.242,P>0.05). Conclusion Levels of IGF-1 and IGFR-1 expres-sion are elevated in prostatic cancer tissue. These findings indicate IGF/IGFR-1 signal pathway could serve to pre-dict the outcome of prostatic cancer.
