目的:利用膜片钳技术阐述益气复脉合剂抗心律失常作用机制。方法采用胶原酶法急性分离大鼠左心室肌细胞,利用膜片钳技术,记录心室肌细胞动作电位。结果异丙肾上腺素(Iso)灌流前,中药组动作电位时程(APD)、APD50、APD90较空白对照组明显延长(P<0.05);Iso灌流后,空白对照组与中药组 APD较灌流前显著延长(P<0.05或P<0.01),空白对照组 APD50、APD90较灌流前亦显著延长(P<0.01);给 Iso后,中药组 APD、APD50、APD90较空白对照组显著缩短(P<0.01)。结论益气复脉合剂可以缩短 Iso引起的 APD的延长,可能与抑制平台期 Ca2+电流和复极后期 K+电流有关。

목적:이용막편겸기술천술익기복맥합제항심률실상작용궤제。방법채용효원매법급성분리대서좌심실기세포,이용막편겸기술,기록심실기세포동작전위。결과이병신상선소(Iso)관류전,중약조동작전위시정(APD)、APD50、APD90교공백대조조명현연장(P<0.05);Iso관류후,공백대조조여중약조 APD교관류전현저연장(P<0.05혹P<0.01),공백대조조 APD50、APD90교관류전역현저연장(P<0.01);급 Iso후,중약조 APD、APD50、APD90교공백대조조현저축단(P<0.01)。결론익기복맥합제가이축단 Iso인기적 APD적연장,가능여억제평태기 Ca2+전류화복겁후기 K+전류유관。
Objective To investigate the anti arrhythmic mechanisms with Yiqi Fumai mixture(YFM) by the patch clamp technique. Methods Using the col agenase method in acutely isolated rat left ventricular myocytes,and using patch clamp techique,to record the action potential of ventricular myocytes.Results Before isoprenaline(Iso)perfusion,compared with the control group,the Chi-nese medicine(CM) group could significantly extend action potential duration(APD),APD50 and APD90 (P<0.05).After Iso perfu-sion,the control group and the CM group could significantly extend APD (P<0.05 or P<0.01),and the control group also could significantly extend APD50 and APD90 (P<0.01).After Iso perfusion,compared with the control group,the CM group could signifi-cantly shortening APD,APD50 and APD90 (P<0.01).Conclusion It suggests that YFM can against the prolongation of APD caused by Iso,maybe related to the inhibiton of the late plateau Ca2+ current and repolarization K+ current.
