目的:寻找一种能诱导多种胚胎干细胞向生殖细胞方向分化的化合物,并以常用的诱导物维甲酸(Retinoic Acid, RA)作为阳性参照物。方法:分别培养小鼠胚胎干细胞1 B10、D3、R1/E,诱导它们形成拟胚体,让拟胚体贴壁生长,加入特定化合物诱导贴壁细胞分化,诱导72 h后,提取被诱导细胞的RNA,再合成cDNA,最后利用实时荧光定量PCR检测各被诱导细胞中与生殖分化相关的基因的表达水平的变化。结果:发现齐墩果酸(Oleanolic Acid,OA)显著上调了所有被研究的小鼠胚胎干细胞的生殖分化关键基因的表达水平,同时发现阳性参照物RA仅能诱导1 B10、R1/E ,而不能诱导D3向生殖细胞方向分化。结论:OA能诱导多种胚胎干细胞向生殖细胞方向分化,诱导能力方面具有比常用诱导物维甲酸更广泛的普遍性。
目的:尋找一種能誘導多種胚胎榦細胞嚮生殖細胞方嚮分化的化閤物,併以常用的誘導物維甲痠(Retinoic Acid, RA)作為暘性參照物。方法:分彆培養小鼠胚胎榦細胞1 B10、D3、R1/E,誘導它們形成擬胚體,讓擬胚體貼壁生長,加入特定化閤物誘導貼壁細胞分化,誘導72 h後,提取被誘導細胞的RNA,再閤成cDNA,最後利用實時熒光定量PCR檢測各被誘導細胞中與生殖分化相關的基因的錶達水平的變化。結果:髮現齊墩果痠(Oleanolic Acid,OA)顯著上調瞭所有被研究的小鼠胚胎榦細胞的生殖分化關鍵基因的錶達水平,同時髮現暘性參照物RA僅能誘導1 B10、R1/E ,而不能誘導D3嚮生殖細胞方嚮分化。結論:OA能誘導多種胚胎榦細胞嚮生殖細胞方嚮分化,誘導能力方麵具有比常用誘導物維甲痠更廣汎的普遍性。
목적:심조일충능유도다충배태간세포향생식세포방향분화적화합물,병이상용적유도물유갑산(Retinoic Acid, RA)작위양성삼조물。방법:분별배양소서배태간세포1 B10、D3、R1/E,유도타문형성의배체,양의배체첩벽생장,가입특정화합물유도첩벽세포분화,유도72 h후,제취피유도세포적RNA,재합성cDNA,최후이용실시형광정량PCR검측각피유도세포중여생식분화상관적기인적표체수평적변화。결과:발현제돈과산(Oleanolic Acid,OA)현저상조료소유피연구적소서배태간세포적생식분화관건기인적표체수평,동시발현양성삼조물RA부능유도1 B10、R1/E ,이불능유도D3향생식세포방향분화。결론:OA능유도다충배태간세포향생식세포방향분화,유도능력방면구유비상용유도물유갑산경엄범적보편성。
Objective:To find an agent which can induce several strains of embryonic stem cells to differentiate towards germ cells,with the common inducer retinoic acid (RA)as positive control.Methods:Different strains of mouse embryonic stem cells (mESC)1 B1 0,D3,R1/E were respectively cultured,and induced to form embryoid bodies (EBs)which were allowed to attach;different compounds were then added to intervene the attached cells;the intervene continued for 72 h and RNA were extracted and cDNA were synthesized;finally,real-time fluorescence quantitative PCR (qPCR)assay were used to measure the transcriptional expression patterns of 1 1 reproductive differentiation related genes.Results:It was found that oleanolic acid (OA)can induce all 3 strains of mouse mESC to differentiate towards germ cells and the positive control RA can only induce mESC-1 B1 0 and mESC-R1/E to differentiate towards germ cells.RA cannot induce mESC-D3 to differentiate towards germ cells.Conclusion:OA can induce several strains of embryonic stem cells to differentiate towards germ cells and it has broader universality than the positive control RA .
