Cholangitis%model construction%rats
目的:探讨建立非活动限制、可复性大鼠急性重症胆管炎(SAC)模型。方法:26只大鼠随机分为SAC造模组和假手术组,两组均进行颈外静脉置管建立无拘束模型,采用胆总管置入麻醉导管,远端结扎,导管内注入内毒素后封管留置。SAC组于胆总管结扎后0、24、48、72 h监测胆总管压力;分别于胆总管注射内毒素后0、12、24、48 h采静脉血检测TBIL、ALT、TNF-α;术后24 h、72 h各取肝脏行病理学检查。结果:SAC组随着造模时间的延长,胆总管压力逐渐提高,不同时点间存在显著性差异(P<0.05);血清TBIL、ALT,TNF-α水平逐渐上升,不同时点均明显高于假手术组相应时间点(P<0.05);病理学检查证实肝脏病理改变明显。结论:新型大鼠SCA模型能够模拟临床,基本能反映人SAC的早期病理过程,也适合SAC术后进一步的实验性研究。

목적:탐토건립비활동한제、가복성대서급성중증담관염(SAC)모형。방법:26지대서수궤분위SAC조모조화가수술조,량조균진행경외정맥치관건립무구속모형,채용담총관치입마취도관,원단결찰,도관내주입내독소후봉관류치。SAC조우담총관결찰후0、24、48、72 h감측담총관압력;분별우담총관주사내독소후0、12、24、48 h채정맥혈검측TBIL、ALT、TNF-α;술후24 h、72 h각취간장행병이학검사。결과:SAC조수착조모시간적연장,담총관압력축점제고,불동시점간존재현저성차이(P<0.05);혈청TBIL、ALT,TNF-α수평축점상승,불동시점균명현고우가수술조상응시간점(P<0.05);병이학검사증실간장병리개변명현。결론:신형대서SCA모형능구모의림상,기본능반영인SAC적조기병리과정,야괄합SAC술후진일보적실험성연구。
Objective To establish a novel rat model of severe acute cholangitis(SAC). Methods Twen?ty-six rats were randomly divided into two groups: model group and shem operated group. All rats were unre?strained and performed with epidural anesthesia catheter used as the filling materials into vena jugularis externa. SAC models were established by placing epidural anesthesia catheter into common bile duct and ligating the dis?tal bile duct, then inside injecting LPS and closing the catheter. The level of common bile duct pressure was measured at 0,24,48,72 h after the ligation. The changes of TBIL,ALT,TNF-α were observed at 0,12,24,48 h af?ter the injection. Pathological changes of liver tissues were observed. Results The level of common bile duct pressure increased gradually after the ligation. Serum TBIL,ALT,TNF-αlevel in model group were progressive in?creased and higher than shem operated group(P<0.05). Marked liver pathological changes were observed in model group. Conclusion This model can reflect the early course of SAC and be used in the further studies of SAC postoperative.
