目的:探讨和研究丙烯酰胺对成年小鼠海马成体神经发生及GSK3β表达的影响。方法:雄性成年昆明小鼠分为丙烯酰胺处理组和生理盐水对照组,通过腹腔注射( i.p.)进行丙烯酰胺染毒,用BrdU标记和免疫组织化学方法研究齿状回神经干细胞的增殖,BrdU/NeuN/GFAP三标的方法研究齿状回神经干细胞的存活和分化,Neuro-2a 细胞中观察GSK3β表达与分布,用Western blot方法检测小鼠海马中GSK3β的表达。结果:丙烯酰胺处理组小鼠齿状回亚颗粒细胞层( SGZ)中的BrdU阳性细胞数量显著低于对照组,并且新生细胞向神经元分化的数量显著降低。丙烯酰胺处理小鼠SGZ区新生干细胞的存活率显著低于对照小鼠,另外,丙烯酰胺组新生干细胞向胶质细胞分化的比例显著升高。在Neuro-2a细胞中磷酸化的GSK3β表达量极显著升高,但在丙烯酰胺处理组小鼠的海马中磷酸化的GSK3β显著降低。结论:结果表明,丙烯酰胺能显著抑制小鼠齿状回成体神经干细胞增殖,降低新生干细胞的存活率并影响其分化,丙烯酰胺抑制小鼠成体神经发生可能与GSK3β有一定的关系。
目的:探討和研究丙烯酰胺對成年小鼠海馬成體神經髮生及GSK3β錶達的影響。方法:雄性成年昆明小鼠分為丙烯酰胺處理組和生理鹽水對照組,通過腹腔註射( i.p.)進行丙烯酰胺染毒,用BrdU標記和免疫組織化學方法研究齒狀迴神經榦細胞的增殖,BrdU/NeuN/GFAP三標的方法研究齒狀迴神經榦細胞的存活和分化,Neuro-2a 細胞中觀察GSK3β錶達與分佈,用Western blot方法檢測小鼠海馬中GSK3β的錶達。結果:丙烯酰胺處理組小鼠齒狀迴亞顆粒細胞層( SGZ)中的BrdU暘性細胞數量顯著低于對照組,併且新生細胞嚮神經元分化的數量顯著降低。丙烯酰胺處理小鼠SGZ區新生榦細胞的存活率顯著低于對照小鼠,另外,丙烯酰胺組新生榦細胞嚮膠質細胞分化的比例顯著升高。在Neuro-2a細胞中燐痠化的GSK3β錶達量極顯著升高,但在丙烯酰胺處理組小鼠的海馬中燐痠化的GSK3β顯著降低。結論:結果錶明,丙烯酰胺能顯著抑製小鼠齒狀迴成體神經榦細胞增殖,降低新生榦細胞的存活率併影響其分化,丙烯酰胺抑製小鼠成體神經髮生可能與GSK3β有一定的關繫。
목적:탐토화연구병희선알대성년소서해마성체신경발생급GSK3β표체적영향。방법:웅성성년곤명소서분위병희선알처리조화생리염수대조조,통과복강주사( i.p.)진행병희선알염독,용BrdU표기화면역조직화학방법연구치상회신경간세포적증식,BrdU/NeuN/GFAP삼표적방법연구치상회신경간세포적존활화분화,Neuro-2a 세포중관찰GSK3β표체여분포,용Western blot방법검측소서해마중GSK3β적표체。결과:병희선알처리조소서치상회아과립세포층( SGZ)중적BrdU양성세포수량현저저우대조조,병차신생세포향신경원분화적수량현저강저。병희선알처리소서SGZ구신생간세포적존활솔현저저우대조소서,령외,병희선알조신생간세포향효질세포분화적비례현저승고。재Neuro-2a세포중린산화적GSK3β표체량겁현저승고,단재병희선알처리조소서적해마중린산화적GSK3β현저강저。결론:결과표명,병희선알능현저억제소서치상회성체신경간세포증식,강저신생간세포적존활솔병영향기분화,병희선알억제소서성체신경발생가능여GSK3β유일정적관계。
Objective:To investigate the influence of acrylamide ( ACR) on adult neurogenesis and expression of GSK 3βin mouse.Methods:Method Adult male Kunming mice were used and divided into two groups:control group and experimental poisoning groups,that were exposured to acrylamide by intraperitoneal injection.Brdu labeling and immunohistochemistry were used to investigate the proliferation of adult neural stem cells in the subgranular zone ( SGZ).BrdU/NeuN/GFAP triple labeling to investigate the survival and differentiation of newly generated cells.Detecting GSK3βexpression and distribution in Neuro-2a cells,the expression of GSK3βwas examined by using Western blot.Results:Compared with control mice ,lower number of BrdU-positive cells and less differentiated into neurons in ACR mice.Less neural stem cells survived ,but more glia cells were generated in the subgranular zone of acrylamide mice.Moreover,higher phosphorylated GSK 3β( Ser9 ) were detected in Neuro-2a cells and mouse dentate gyrus in ACR mice respectively .Conclusion:These results suggested that acrylamide inhibits neural stem cells proliferation and influences the survival and differentiation of newly generated cells.Acrylamide inhibits neurogenesis maybe through GSK 3βsignaling pathway.
