利用超声耦合乙醇-硫酸铵双水相萃取技术提取牛大力总黄酮。通过单因素实验和正交实验对影响牛大力总黄酮提取的料液比、硫酸铵浓度、乙醇浓度和超声时间等因素进行探讨,以建立牛大力总黄酮的提取工艺。结果显示:影响牛大力总黄酮提取因素的主次顺序为料液比>乙醇浓度>硫酸铵浓度>超声时间;最优提取条件为乙醇浓度80%、料液比1∶40(m∶V )、超声时间50 min、硫酸铵浓度为0.3 g/mL,在此条件下,牛大力总黄酮得率为5.82 mg/g。

In this study, flavonoids from Millettia speciosa roots was extracted by ultrasonic-assisted alcohol-ammonium sulphate aqueous two-phase system. To determine the optimal extraction conditions, the effect of the conditions on yield, such as the ethanol concentration, the solid-liquid ratio, the amount of the ammonium sulfate and the ultrasonic time were studied by single factor experiment and orthogonal test. The results showed that the order of factor affecting total flavones extraction was solid-liquid ratio >ethanol concentration > ammonium sulfate concentration > ultrasonic time. The optimum extracting conditions were as follows: the volume fraction of alcohol 80%, solid to solution ratio of 1:40, ultrasonic time 50 min, the ammonium sulfate 0.3 g/mL. At the optimum conditions, the extraction rate of the flavonoids could reach 5.82 mg/g.
