目的：研究莲房原花青素（LSPC）对乙醇诱导的人胚肝细胞株L－02氧化损伤的保护作用。方法观察200mmol? L －1乙醇，5、10、25mg? L －1的LSPC，5、10、25mg? L －1的LSPC与200mmol? L －1乙醇共同作用于肝细胞 L－0224h 后，运用单细胞凝胶电泳（ SCGE）实验和微核实验（MNT）观察DNA和染色体损伤情况，用流式细胞术（FCM）分析凋亡率的变化以及活性氧探针（2，7－二氯荧光素）探测细胞内活性氧（ROS）水平的变化。结果 LSPC组与正常对照组比较，差异无显著性意义（ P＞0．05）。纯乙醇组与正常对照组比较，差异有显著性意义，与乙醇组相比，5mg? L －1和10mg? L －1 LSPC能显著降低olive尾矩（OTM）、微核率（MNF）、凋亡率和ROS的水平（均P＜0．05），但高剂量组（25mg? L －1）对乙醇引起的肝细胞损伤保护作用不明显。结论适量浓度的莲房原花青素对乙醇诱导的肝细胞 L－02的氧化损伤有保护作用。其机制可能与LSPC的抗氧化活性有关。
목적：연구련방원화청소（LSPC）대을순유도적인배간세포주L－02양화손상적보호작용。방법관찰200mmol? L －1을순，5、10、25mg? L －1적LSPC，5、10、25mg? L －1적LSPC여200mmol? L －1을순공동작용우간세포 L－0224h 후，운용단세포응효전영（ SCGE）실험화미핵실험（MNT）관찰DNA화염색체손상정황，용류식세포술（FCM）분석조망솔적변화이급활성양탐침（2，7－이록형광소）탐측세포내활성양（ROS）수평적변화。결과 LSPC조여정상대조조비교，차이무현저성의의（ P＞0．05）。순을순조여정상대조조비교，차이유현저성의의，여을순조상비，5mg? L －1화10mg? L －1 LSPC능현저강저olive미구（OTM）、미핵솔（MNF）、조망솔화ROS적수평（균P＜0．05），단고제량조（25mg? L －1）대을순인기적간세포손상보호작용불명현。결론괄량농도적련방원화청소대을순유도적간세포 L－02적양화손상유보호작용。기궤제가능여LSPC적항양화활성유관。
ABSTRACT:OBJECTIVE To study the protective effect of lotus seedpod procyanidns ( LSPC ) on ethanol-in-duced oxidative damage to human embryo liver L-02 cell.METHODS L-02 cells were exposed to 200 mmol? L-1 ethanol, 5、10 and 25mg? L-1 LSPC, 200mmol? L-1 ethanol +5、10 and 25mg? L-1 LSPC respectively for 24hrs.The single cell gel electrophoresis (SCGE)and micronucleus test (MNT) were used to detect the breakage of DNA strand and chromosome breaks .The changes of the apoptosis rates and the level of reactive oxygen species (ROS) were determined by flow cytometry ( FCM) and a probe (2,7-dichloro fluoresceinciactate ,DCFH-DA) re-spectively.RESULTS There were no significant differences between LSPC and control group .The difference being significant as ethanol group compared with the normal control group (P<0.05).Compared with the ethanol group,5 and 10mg? L-1 LSPC group decreased the Olive tail moment (OTM),the micronuclei frequencies (MNF),the ap-optosis rate and ROS level ( P <0.05 ) .But the same effect didn ’ t be observed in the 25mg? L-1 LSPC group.CONCLUSION LSPC with appropriate concentration can protect ethanol induced damage effect in human embryo liver L-02 cells.The antioxidant of LSPC may be the underlying mechanism .
