目的:探讨姜黄素+5-FU对粘液表皮样癌MEC-1的抑制作用。方法:复苏MEC-1细胞株,进行传代培养,当细胞达到对数生长期时进行分组:实验组为姜黄素组、5-FU组、姜黄素+5-FU组;对照组为PBS组;在MTT实验中,实验组、对照组均由低到高分别设6种不同浓度:5μmol/L、10μmol/L、20μmol/L、40μmol/L、80μmol/L、160μmol/L,观察其对MEC-1细胞株的体外生长活性的影响;在流式细胞术分析实验中,实验组、对照组分别设3种适中浓度:20μmol/L、40μmol/L、80μmol/L,观察其对肿瘤细胞的细胞周期及凋亡率的影响。结果:MTT法观察到6种不同浓度的实验组对肿瘤细胞MEC-1均有明显抑制作用,且这种抑制作用与浓度和作用时间成正相关;流式细胞术分析结果表明,对于MEC-1的细胞周期及凋亡率,3种不同浓度的实验组均能产生影响,且随着浓度的递增,出现G0/G1期随之下降,S期无明显变化,G2/M期细胞随之上升的表现;细胞凋亡率变化表明,姜黄素+5-FU比单独组更能有效诱导肿瘤细胞MEC-1凋亡。结论:姜黄素与5-FU均能有效抑制粘液表皮样癌MEC-1的生长并使其凋亡,且姜黄素能增强5-FU对MEC-1细胞的抑制效果。

목적:탐토강황소+5-FU대점액표피양암MEC-1적억제작용。방법:복소MEC-1세포주,진행전대배양,당세포체도대수생장기시진행분조:실험조위강황소조、5-FU조、강황소+5-FU조;대조조위PBS조;재MTT실험중,실험조、대조조균유저도고분별설6충불동농도:5μmol/L、10μmol/L、20μmol/L、40μmol/L、80μmol/L、160μmol/L,관찰기대MEC-1세포주적체외생장활성적영향;재류식세포술분석실험중,실험조、대조조분별설3충괄중농도:20μmol/L、40μmol/L、80μmol/L,관찰기대종류세포적세포주기급조망솔적영향。결과:MTT법관찰도6충불동농도적실험조대종류세포MEC-1균유명현억제작용,차저충억제작용여농도화작용시간성정상관;류식세포술분석결과표명,대우MEC-1적세포주기급조망솔,3충불동농도적실험조균능산생영향,차수착농도적체증,출현G0/G1기수지하강,S기무명현변화,G2/M기세포수지상승적표현;세포조망솔변화표명,강황소+5-FU비단독조경능유효유도종류세포MEC-1조망。결론:강황소여5-FU균능유효억제점액표피양암MEC-1적생장병사기조망,차강황소능증강5-FU대MEC-1세포적억제효과。
Objective:To investigate the inhibitory effect of curcumin and 5-FU on MEC-1 cells of mucoepidermoid carcinoma. Methods: MEC-1 cells were revived and cultured, cells reached a logarithmic growth phase were divided in experimental group, including curcumin group, 5-FU group, curcumin and 5-FU group, and control group which is PBS group; experimental group and control group were arranged concentration of 5μmol/L, 10μmol/L, 20μmol/L, 40μmol/L, 80μmol/L, 160μmol/L from low to high in the MTT experiment, to observe the effects on vitro growth ac-tivity of MEC-1 cells; in the flow cytometry analysis experiment, experimental group and control group were arranged moderate concentration of 20μmol/L, 40μmol/L, 80μmol/L, to observe the effects on cell cycle and apoptosis rate of tumor cells. Results:6 different concentration of experi-mental groups had obviously inhibitory effects on tumor cells and positive correlation with concentration and action time in MTT experiment;flow cytometry analysis showed that 3 different concentration of experimental groups had effects on cell cycle and apoptosis rate of MEC-1,and with the concentration increasing, G0/G1 phase decreased, S phase without significant changes, G2/M phase cells increased;apoptosis rate changes indicated that curcumin and 5-FU could induce the apoptosis of MEC-1 tumor cells more effectively than single group. Conclusion:Curcumin and 5-FU can ef-fectively inhibit the growth of mucoepidermoid carcinoma MEC-1 and make them apoptosis, and curcumin can enhance the inhibitory effect of 5-FU on MEC-1 cells.
