目的:近来研究发现过敏和自身免疫性病变之间或许存在联系,嗜碱性粒细胞和组织肥大细胞在哮喘的病理过程中发挥作用。本研究的目的是在儿童哮喘患者中检测FcεRI功能性自身抗体的对嗜碱性粒细胞的反应性,通过运用流式细胞嗜碱性粒细胞活化试验测定CD63的表达。方法从2012年1月~2014年6月,随机纳入于厦门市妇幼保健院儿科病房就诊,根据美国胸科协会制定的诊断指南诊断为哮喘的儿童,非过敏的健康志愿者的血清刺激患者组以及对照组以后,采用基于流式细胞仪的嗜碱性粒细胞活化试验检测方法,测定CD63的表达,因CD63的表达可用于识别嗜碱性粒细胞是否被患者或者健康对照组的血清激活。结果共纳入研究的哮喘儿童有72名,健康对照组32例。29/78(37.2%)的哮喘患者血清表达CD63+的嗜碱性粒细胞,而在健康受试者的血清中只有4/32(12.5%)表达CD63+嗜碱性粒细胞,在哮喘患者中CD63+嗜碱性粒细胞的比例较健康受试者相比差异具有统计学意义(P<0.05)。结论 FcεRI自身抗体可能与儿童哮喘的疾病相关。

목적:근래연구발현과민화자신면역성병변지간혹허존재련계,기감성립세포화조직비대세포재효천적병리과정중발휘작용。본연구적목적시재인동효천환자중검측FcεRI공능성자신항체적대기감성립세포적반응성,통과운용류식세포기감성립세포활화시험측정CD63적표체。방법종2012년1월~2014년6월,수궤납입우하문시부유보건원인과병방취진,근거미국흉과협회제정적진단지남진단위효천적인동,비과민적건강지원자적혈청자격환자조이급대조조이후,채용기우류식세포의적기감성립세포활화시험검측방법,측정CD63적표체,인CD63적표체가용우식별기감성립세포시부피환자혹자건강대조조적혈청격활。결과공납입연구적효천인동유72명,건강대조조32례。29/78(37.2%)적효천환자혈청표체CD63+적기감성립세포,이재건강수시자적혈청중지유4/32(12.5%)표체CD63+기감성립세포,재효천환자중CD63+기감성립세포적비례교건강수시자상비차이구유통계학의의(P<0.05)。결론 FcεRI자신항체가능여인동효천적질병상관。
Objective Recent studies have found an association between autoimmune diseases and allergies may, basophils and tissue mast cells play a role in the pathogenesis of asthma. The purpose of this study is to test the functionality FcεRI autoantibodies in children with asthma against basophil reactivity, using the expression of CD63 was determined by flow cytometry basophil activation assay. Methods From January 2012 to June 2014,in the pediatric ward randomly into treatment of maternal and Child Health Hospital of Xiamen City, the asthma diagnosis was made according to American Thoracic Society diagnostic guidelines, after the stimulation of serum from non-allergic donors to patients and controlling groups, using lfow cytometry based basophil activation assay method to detection expression of CD63, expression of CD63 can be help to make sure whether the basophils cells are activated in patient, or healthy controls. Results The study included a total of 72 children with asthma, healthy control group of 32 patients. 29/78 (37.2%) in the serum of patients with asthma expression of CD63+basophils, and in the serum of healthy subjects, only 4/32 (12.5%) the expression of CD63+basophils in patients with asthma CD63+percentage of basophils compared statistically significant (P<0.05) compared with healthy subjects. Conclusion FcεRI autoantibodies may be disease-related asthma in children.
