目的:研究红花甙(carthamin)对过氧化氢(H2O2)诱导神经母细胞瘤 SH-SY5Y 细胞(SH-SY5Y cells)凋亡的影响,并探讨其机制。方法实验分为空白对照组、H2O2损伤组及红花甙高、中、低剂量组。用H2O2作用于SH-SY5Y细胞,使其发生凋亡。 MTT法检测SH-SY5Y细胞活性,比色法测丙二醛(MDA)的含量,流式细胞仪检测细胞凋亡。结果与损伤组比较,随着红花甙剂量的增加,SH-SY5Y细胞存活率提高(P<0.01),MDA生成量减少(P<0.01),凋亡率下降(P<0.05)。结论红花甙对H2O2诱导SH-SY5Y细胞凋亡有保护作用,与红花甙能抗氧化、降低细胞凋亡有关。

목적:연구홍화대(carthamin)대과양화경(H2O2)유도신경모세포류 SH-SY5Y 세포(SH-SY5Y cells)조망적영향,병탐토기궤제。방법실험분위공백대조조、H2O2손상조급홍화대고、중、저제량조。용H2O2작용우SH-SY5Y세포,사기발생조망。 MTT법검측SH-SY5Y세포활성,비색법측병이철(MDA)적함량,류식세포의검측세포조망。결과여손상조비교,수착홍화대제량적증가,SH-SY5Y세포존활솔제고(P<0.01),MDA생성량감소(P<0.01),조망솔하강(P<0.05)。결론홍화대대H2O2유도SH-SY5Y세포조망유보호작용,여홍화대능항양화、강저세포조망유관。
Objective To research for effect of carthamin on H2O2 induced neuroblastoma SH-SY5Y cells apoptosis,and to explore its mechanism. Methods Experimental cell strains were divided into blank group,H2O2 injury group as well as carthamin high,medium and low dose group. By using H2O2 to make SH-SY5Y cells apoptosis was taken. MTT method was used to detect SH-SY5Y cell activity,colorimetric method to measure malondialdehyde (MDA) content and flow cytometry to detect cell apoptosis. Results Compared with injury group,with the increase of carthamin dose,SH-SY5Y cell survival rate increased,MDA concentration decreased and the rate of apoptosis decreased. Conclusion Carthamin could protect SH-SY5Y cells from H2O2 injury which would be associated with anti-oxidation and anti-apoptosis of carthamin.
