为获取γ-PGA高产突变株,确定聚谷氨酸对钙溶解性和对小鼠体内钙吸收的影响。使用紫外照射纳豆芽孢杆菌诱导突变,根据形态特征变化筛选、培养突变株。分离纯化不同突变株的γ-聚谷氨酸(γ-PGA)并测定产量。筛选出γ-PGA高产的突变株并测定其遗传稳定性。测定γ-PGA在体外和体内对钙溶解性的影响以及γ-PGA对小鼠骨矿物质含量的影响。筛选出了具有稳定遗传性的γ-PGA高产突变株Z-29,其γ-PGA的产量大于18 g/L。结果表明,在体外,γ-PGA能促进钙的溶解性;在体内,γ-PGA也能促进小鼠小肠钙的吸收,并能增加小鼠骨钙和骨磷的含量。筛选出了纳豆芽孢杆菌γ-PGA高产的突变株,证明了γ-PGA在体内外都能增加钙的溶解性,这为更深入地研究γ-PGA的功能和生产应用提供了依据。

The purpose of this study is to obtain mutant strain with high yields of γ-PGA and determine polyglutamic acid effect on calcium absorption in mice .Bacillus natto was irradiated by ultraviolet to induce muta-tion.The mutant strains were selected according to morphological changes .Then,the mutant strains were cultivated . The γ-poly glutamic acid (γ-PGA) of different mutant strains were separated and purified to determine its produc-tion.The mutant strain which had high yields of γ-PGA was selected and determined its genetic stability .The effects of the calcium solubility by γ-PGA in vitro and in vivo were measured as well as the bone mineral content in mice . Experiment screened out a mutant strain ,Z-29,with stable high yields of γ-PGA greater than 18 g/L.The experi-mental results showed that γ-PGA promoted calcium solubility in vitro and in mice small intestine .Moreover,it in-creased bone calcium and bone phosphorus content in mice .This experiment gained Bacillus natto mutant strains with high yields of γ-PGA and proved that γ-PGA can increase the solubility of calcium in vitro and in vivo.It pro-vided the basis for further research and application in production .
