目的:观察蒺藜提取物对小鼠热损伤睾丸一氧化氮合酶(NOS)-一氧化氮(NO)系统的影响,探讨蒺藜对小鼠热损伤睾丸的保护修复作用机制。方法健康雄性小鼠30只,随机分为对照组、模型组和给药组,每组10只;建立睾丸热损伤模型,给予治疗剂量的蒺藜提取物;睾丸NOS活力和NO含量测定;睾丸NOS和caspase-3免疫组织化学染色,光学显微镜观察;酶联法检测血清中睾酮含量。结果模型组与对照组相比睾丸组织NOS活力和NO含量明显升高,睾丸iNOS阳性表达明显增加,睾丸caspase-3阳性细胞明显增多,血清睾酮含量明显降低。给药组与模型组相比睾丸组织NOS活力和NO含量明显降低,睾丸iNOS阳性表达明显减少,睾丸caspase-3阳性细胞明显减少,血清睾酮含量明显升高。结论蒺藜提取物通过调控NOS-NO系统,实现对小鼠热损伤睾丸的保护修复作用。

목적:관찰질려제취물대소서열손상고환일양화담합매(NOS)-일양화담(NO)계통적영향,탐토질려대소서열손상고환적보호수복작용궤제。방법건강웅성소서30지,수궤분위대조조、모형조화급약조,매조10지;건립고환열손상모형,급여치료제량적질려제취물;고환NOS활력화NO함량측정;고환NOS화caspase-3면역조직화학염색,광학현미경관찰;매련법검측혈청중고동함량。결과모형조여대조조상비고환조직NOS활력화NO함량명현승고,고환iNOS양성표체명현증가,고환caspase-3양성세포명현증다,혈청고동함량명현강저。급약조여모형조상비고환조직NOS활력화NO함량명현강저,고환iNOS양성표체명현감소,고환caspase-3양성세포명현감소,혈청고동함량명현승고。결론질려제취물통과조공NOS-NO계통,실현대소서열손상고환적보호수복작용。
Objective To investigate the effects of Tribulus Terrestris extract on the NOS-NO system of the thermal-injured testis in mice, and explore its mechanism.Methods All 30 healthy male mice were randomly divided into the control group, the model group and the treated group, 10 mice per group. The thermal-injured testis models of mice were established and then were administrated with dose of Tribulus terrestris extract. NOS activity and NO content of the testis were measured. Levels of testicular NOS and Caspase-3 were detected by immunohistochemistry. The content of testosterone in serum was detected by ELISA.Results Compared with that of the control group, activities of NOS and the content of NO in testicular tissues were increased significantly in the model group. The positive expression of iNOS and the number of testicular Caspase-3 positive cells were also increased obviously, but the serum concentration of testosterone was decreased. Compared with that of the model group, activities of NOS and the content of NO of the testicular tissue were decreased in the treated group. The positive expression of iNOS and the number of testicular Caspase-3 positive cells were also decreased significantly, but the serum concentration of testosterone was increased. Conclusion Tribulus terrestris extract shows protect effects on the thermal-injuried testis in mice by regulating the NOS-NO system.
