目的:探索长链非编码RNA-HOTAIR在乳腺癌及前哨淋巴结、腋窝淋巴结中的表达情况,分析与乳腺癌其他指标的相关关系,以指导治疗并预测预后。方法:收集2013年6月-2013年12月乳腺癌住院手术患者10例,均行前哨淋巴结活检术,取乳腺肿块组织、癌旁组织及前哨淋巴结(SLN)、腋窝淋巴结(ALN)。术后均经病理证实为浸润性导管癌。四组标本组织采用实时荧光定量RT-PCR检测HOTAIR的表达情况,结合临床病理结果,采用t检验、等级相关等统计方法分析各指标之间的关系。结果:(1)HOTAIR基因在各组中的表达,SLN组(9.33±1.23),ALN组(6.90±2.00),两者比较差异有统计学意义(t=3.401,P=0.004);癌组织组(8.81±1.27),癌旁组织组(7.00±1.11),两者比较差异有统计学意义(t=3.389,P=0.003);SLN组与癌旁组织组比较差异有统计学意义(t=4.441,P<0.001)。(2)HOTAIR表达情况与临床病理指标关系密切,与淋巴结转移率、ki-67蛋白表达呈正相关,相关系数分别为0.67和0.82,差异有统计学意义(P<0.05)。结论:HOTAIR基因在乳腺癌前哨淋巴结以及癌组织中有异常高表达,其促进细胞增殖、侵袭转移的功能显著,提供乳腺癌基因靶向治疗的新位点。

목적:탐색장련비편마RNA-HOTAIR재유선암급전초림파결、액와림파결중적표체정황,분석여유선암기타지표적상관관계,이지도치료병예측예후。방법:수집2013년6월-2013년12월유선암주원수술환자10례,균행전초림파결활검술,취유선종괴조직、암방조직급전초림파결(SLN)、액와림파결(ALN)。술후균경병리증실위침윤성도관암。사조표본조직채용실시형광정량RT-PCR검측HOTAIR적표체정황,결합림상병리결과,채용t검험、등급상관등통계방법분석각지표지간적관계。결과:(1)HOTAIR기인재각조중적표체,SLN조(9.33±1.23),ALN조(6.90±2.00),량자비교차이유통계학의의(t=3.401,P=0.004);암조직조(8.81±1.27),암방조직조(7.00±1.11),량자비교차이유통계학의의(t=3.389,P=0.003);SLN조여암방조직조비교차이유통계학의의(t=4.441,P<0.001)。(2)HOTAIR표체정황여림상병리지표관계밀절,여림파결전이솔、ki-67단백표체정정상관,상관계수분별위0.67화0.82,차이유통계학의의(P<0.05)。결론:HOTAIR기인재유선암전초림파결이급암조직중유이상고표체,기촉진세포증식、침습전이적공능현저,제공유선암기인파향치료적신위점。
Objective:To explore the expression of long chain of non-encoded RNA-HOTAIR in sentinel lymph node and axillary lymph node in breast cancer, to analyze the correlation with other indicators of breast cancer, and to help guide treatment and predict outcome.Method: Data from 10 surgical patients of breast cancer hospitalized from June 2013 to December 2013 was collected. Sentinel lymph node biopsy was performed, and breast tumor tissue, paracancerous tissue,Sentinel lymph node(SLN),and axillary lymph nodes (ALN) were collected.Invasive ductal carcinoma was confirmed by pathological examination.Expression of HOTAIR in 4 sets of specimen tissue was measured by real-time fluorescence quantitative RT-PCR.Combined with clinical pathology, the correlation with other indicators was evaluated usingt test and rank correlation.Result:(1)HOTAIR gene expression in each group was measured.(9.33±1.23) for SLN group and (6.90±2.00) for ALN Group (t=3.401,P=0.004).(8.81±1.27) for Cancer Group, and (7.00±1.11) for paracancerous tissue group (t=3.389,P=0.003).The difference of the expression of HOTAIR between SLN group and the adjacent tissue Group was statistically significant(t=4.441,P<0.001).(2)HOTAIR expression was correlated with Clinical pathological indicators. It was positively correlated with Ki-67 protein expression and lymph node metastasis rate, and the correlation coefficient of 0.67 and 0.82, respectively(P<0.05).Conclusion:Abnormal high expression of HOTAIR was observed in the Sentinel Lymph nodes of breast cancer as well as the tumor tissue.It can promote cell proliferation, invasion and metastasis,which may provide new therapeutic target for breast cancer.
