Aconite%black prepared lateral root of aconite%producing area%acute heart failure
目的:研究不同产地四川江油附子、四川凉山附子、云南禄劝附子、陕西汉中附子对急性心衰大鼠心功能指标的影响及其作用机制。方法:用戊巴比妥钠建立大鼠急性心衰模型,各组均以给药前各项指标对照,16道生理记录仪同步记录左室内压(LVSP),左室内压最大上升速率(+dp/dtmax),左室内压最大下降速率(-dp/dtmin),动态观察不同产地附子醇提物(5 g·kg-1)对急性心衰大鼠心功能指标及血浆中精氨酸加压素(AVP)、内皮素-1(ET-1)、心钠素(ANP)、血管紧张素(A域)水平变化及脑钠素(BNP)基因合成与表达的影响。结果:不同产地附子均能升高急性心衰大鼠左室内压,提高左心室+dp/dtmax和-dp/dtmin;并能降低心衰大鼠血浆中AVP水平含量、抑制心室肌内BNP基因的合成和表达,与抑制缩血管因子ET-1、A域水平及ANP基因表达无关。结论:不同产地附子炮制品均能改善大鼠心功能指标,其作用机制与左室功能抑制,心室肌收缩有关;对血管收缩导致的心脏后负荷增加有抑制作用,对前负荷增加无影响。并以道地产区江油附子作用最为明显,表明以功效为核心的中药质量评价模式来评价药材品质、控制药材质量的可行性。

목적:연구불동산지사천강유부자、사천량산부자、운남록권부자、협서한중부자대급성심쇠대서심공능지표적영향급기작용궤제。방법:용무파비타납건립대서급성심쇠모형,각조균이급약전각항지표대조,16도생리기록의동보기록좌실내압(LVSP),좌실내압최대상승속솔(+dp/dtmax),좌실내압최대하강속솔(-dp/dtmin),동태관찰불동산지부자순제물(5 g·kg-1)대급성심쇠대서심공능지표급혈장중정안산가압소(AVP)、내피소-1(ET-1)、심납소(ANP)、혈관긴장소(A역)수평변화급뇌납소(BNP)기인합성여표체적영향。결과:불동산지부자균능승고급성심쇠대서좌실내압,제고좌심실+dp/dtmax화-dp/dtmin;병능강저심쇠대서혈장중AVP수평함량、억제심실기내BNP기인적합성화표체,여억제축혈관인자ET-1、A역수평급ANP기인표체무관。결론:불동산지부자포제품균능개선대서심공능지표,기작용궤제여좌실공능억제,심실기수축유관;대혈관수축도치적심장후부하증가유억제작용,대전부하증가무영향。병이도지산구강유부자작용최위명현,표명이공효위핵심적중약질량평개모식래평개약재품질、공제약재질량적가행성。
This article was aimed to study influence on heart function indexes and its mechanism of aconite from Jiangyou in Sichuan province, Liangshan in Sichuan province, Luquan in Y unnan province, and Hanzhong in Shanxi province among rats with acute heart failure (AHF). The AHF model rat was induced by injecting pentobarbital sodi-um. Parameters including the left ventricular pressure (LVSP), the maximum rate of pressure rise of left ventricular pressure (+dp/dtmax), the largest decline in the rate of left ventricular pressure (-dp/dtmin) were detected with 16 physi-ological recorder. The ethanol extractions of aconite (5 g·kg-1) with different origins were administered to AHF rats to observe the influence on heart function indexes and concentration changes of AVP, ET-1, ANP, AII, and the syn-thesis and expression of BNP gene. The results showed that aconite from different origins can increase the LVSP,+dp/dtmax, -dp/dtmin of AHF rats. It can also decrease the concentration of AVP, inhibit synthesis and expression of BNP gene. It was irrelevant to the inhibition of vasoconstrictor factor of ET-1, AII and ANP gene expression. It was concluded that processed aconite from different origins can improve the heart function indexes of rats. The mecha-nism was correlated to the inhibition of left ventricular and ventricular contraction. It had inhibiting effect on after-load, but no effect on preload. The effect of aconite from Jiangyou, which was the geo-authentic product area, was the most obvious one. It showed the feasibility of using effect as core Chinese medicine quality evaluation mode in the quality control of Chinese medicine.
