目的:建立并优化整体鸡胚原位杂交的实验方法,降低背景,提高信号强度,为后续实验提供良好条件。方法通过对整体鸡胚Bmp4(bone morphogenetic protein 4,骨形态蛋白)基因的原位杂交,摸索实验过程中起决定作用的影响因素及其最佳工作状态。结果在整体鸡胚Bmp4原位杂交均获得高特异性和高强度的杂交信号,且背景色低。结论成功建立并优化了整体鸡胚原位杂交体系。

목적:건립병우화정체계배원위잡교적실험방법,강저배경,제고신호강도,위후속실험제공량호조건。방법통과대정체계배Bmp4(bone morphogenetic protein 4,골형태단백)기인적원위잡교,모색실험과정중기결정작용적영향인소급기최가공작상태。결과재정체계배Bmp4원위잡교균획득고특이성화고강도적잡교신호,차배경색저。결론성공건립병우화료정체계배원위잡교체계。
Objective To report an experimental protocol for in situ hybridization in whole chick embryo with improved signal specificity and strength and reduced background noise for favorable conditions for future studies. Methods In situ hy?bridization of Bmp4(bone morphogenetic protein 4)in whole chick embryo was conducted and key factors affecting the testing process and optimal working conditions were investigated. Results Hybridization signals of high specificity and strength were obtained for Bmp4 with low background signals with optimized methodologies. Conclusions We have successfully established and optimized in situ hybridization of Bmp4 in chick whole embryo.
