目的:建立大鼠颅骨骨缺损局部持续灌注β‐神经生长因子(β‐NGF)给药模型。方法利用外科手术方法切取SD大鼠颅骨顶部左右两侧5 mm的骨质,建立配对大鼠颅骨缺损,利用Alzet微渗透泵右侧骨缺损区持续灌注含β‐NGF的PBS溶液,左侧仅灌注PBS溶液,持续灌注7 d。术后28 d通过H‐E染色和Gomori改良三色染色观察颅骨缺损的愈合情况,检测建模是否成功。结果实验大鼠未发现术后伤口感染、裂开及微渗透泵脱落丢失现象。H‐E及Gomori改良三色染色结果显示,骨缺损区均可见有新骨的形成和骨矿化。结论在大鼠颅骨骨缺损的基础上,建立利用Alzet微渗透泵行局部持续灌注β‐NGF的给药模型是可行的。

목적:건립대서로골골결손국부지속관주β‐신경생장인자(β‐NGF)급약모형。방법이용외과수술방법절취SD대서로골정부좌우량측5 mm적골질,건립배대대서로골결손,이용Alzet미삼투빙우측골결손구지속관주함β‐NGF적PBS용액,좌측부관주PBS용액,지속관주7 d。술후28 d통과H‐E염색화Gomori개량삼색염색관찰로골결손적유합정황,검측건모시부성공。결과실험대서미발현술후상구감염、렬개급미삼투빙탈락주실현상。H‐E급Gomori개량삼색염색결과현시,골결손구균가견유신골적형성화골광화。결론재대서로골골결손적기출상,건립이용Alzet미삼투빙행국부지속관주β‐NGF적급약모형시가행적。
Objective To evaluate the effect of nerve growth factor beta(β‐NGF) on bone forma‐tion and bone remodeling ,topical continuous infusion of β‐NGF in rat skull critical bone defects was estab‐lished . Methods Two 5 mm diameter bone defects were symmetrically created surgically on the parietal bone . The bone defect on the right side was given by β‐NGF PBS solution and the left side was given only by PBS solution via Alzet osmotic pump over 7 days after surgery . New bone formation was evaluated using H‐E staining and Gomori modified trichrome staining at 28 days postoperation . Results All the wounds healed well without infection ,dehiscence and pump lost . According to the H‐E staining and Gomori modified trichrome staining ,new bone formation and bone remodeling were showed after 28 days . Conclusion The animal model of topical continuous infusion of β‐NGF via osmotic pump followed mak‐ing rat skull bone defects were established successfully .
