Eucommia ulmoides%Flavone%Hepatitis Bvirus%Hepatitis Be antigens%Hepatitis Bsurface anti-gens%In vitro
目的:观察杜仲总黄酮在体外抗乙型肝炎病毒(HBV)的效果。方法用乙醇分段法提取杜仲皮中总黄酮。以转染HBV 的人肝癌细胞HepG2.2.15为靶细胞,以药物拉米夫定(50μg/mL)作为阳性对照,以杜仲总黄酮浓度0μg/mL为空白对照组,采用MTT法测定杜仲总黄酮对HepG2.2.15细胞的毒性。采用Real-time PCR技术和酶联免疫吸附试验(ELISA)检测不同剂量(25、50、100、200μg/mL)杜仲总黄酮体外抗HBV的作用。结果杜仲总黄酮各剂量组的OD值均低于空白对照组,其中50、100、200μg/mL剂量组抑制率与空白对照组比较,差异均有统计学意义(P<0.05)。杜仲总黄酮各剂量组对HBV-DNA的复制和乙型肝炎e抗原(HBeAg)、乙型肝炎表面抗原(HBsAg)的分泌均有抑制作用,与空白对照组比较,差异均有统计学意义(P<0.05)。结论杜仲总黄酮在体外有抗HBV作用。

목적:관찰두중총황동재체외항을형간염병독(HBV)적효과。방법용을순분단법제취두중피중총황동。이전염HBV 적인간암세포HepG2.2.15위파세포,이약물랍미부정(50μg/mL)작위양성대조,이두중총황동농도0μg/mL위공백대조조,채용MTT법측정두중총황동대HepG2.2.15세포적독성。채용Real-time PCR기술화매련면역흡부시험(ELISA)검측불동제량(25、50、100、200μg/mL)두중총황동체외항HBV적작용。결과두중총황동각제량조적OD치균저우공백대조조,기중50、100、200μg/mL제량조억제솔여공백대조조비교,차이균유통계학의의(P<0.05)。두중총황동각제량조대HBV-DNA적복제화을형간염e항원(HBeAg)、을형간염표면항원(HBsAg)적분비균유억제작용,여공백대조조비교,차이균유통계학의의(P<0.05)。결론두중총황동재체외유항HBV작용。
Objective To investigate the effect against hepatitis B virus(HBV) of total flavone eucommia ulmoides in vitro. Methods The total flavone was extracted from eucommia ulmoides skin by alcoholic fractionation method. It was adopted MTT method to detect the toxicity of the total flavone eucommia ulmoides on HepG2.2.15 cells under the human hepatocellular carcino-ma cells HepG2.2.15 cells with HBV transfection as the target cells,the lamivudine(50μg/mL) as the positive control group,and total flavone eucommia ulmoides concertration (0μg/mL) as the blank control group. The anti-HBV activity in vitro at doses of 25μg/mL,50 μg/mL,100 μg/mL,200 μg/mL was detected by Real-time PCR and enzyme linked immunosorbent assay (ELISA). Results The OD value of total flavone eucommia ulmoides at above variuos doses was lower than that of the blank control group , in which,the difference of inhibition rate at the dose of 50μg/mL,100μg/mL,200μg/mL had statistcal significance (P<0.05) compared with the blank control group. The total flavone eucommia ulmoided at above variuos doses had a inhibiting effect on du-plication of HBV-DNA and secretion of HBeAg and HBsAg,which had statistical significance in diffrence(P<0.05). Conclusion Total flavone Eucommia ulmoides has the anti-HBV activity in vitro.
