目的:通过测定与分析各组大鼠血清及肺泡灌洗液(BALF)中γ干扰素(IFN-γ)、白细胞介素4(IL-4)含量,观察黄龙止咳口服液对咳嗽变异性哮喘(CVA)模型大鼠免疫功能的影响,进而探讨黄龙止咳口服液调节CVA免疫功能紊乱的作用机制。方法利用卵蛋白(OA)和氢氧化铝共同激发致敏建立CVA大鼠模型,将大鼠分为正常组、模型组、黄龙高剂量组、黄龙中剂量组、黄龙低剂量组、先声咳喘宁组、布地奈德组,记录引喘诱咳时2 min内大鼠咳嗽次数及血清和支气管BALF中IL-4、IFN-γ含量。结果与正常组比较,模型组咳嗽次数明显增多(P<0.01),血清和支气管BALF中IL-4含量显著升高(P<0.01),IFN-γ含量降低(P<0.01);与模型组比较,各治疗组咳嗽次数均明显减少(P<0.01),血清及BALF中IL-4含量明显降低,IFN-γ含量明显升高,差异有统计学意义(P<0.05或P<0.01);黄龙高剂量组与黄龙低、中剂量组比较,咳嗽次数明显减少,血清及BALF中IL-4含量明显降低,IFN-γ含量明显升高,差异有统计学意义(P<0.05或P<0.01);与先声咳喘宁组比较,黄龙高剂量组血清IFN-γ值相对升高,BALF中IL-4值相对降低,IFN-γ值相对升高,差异有高度统计学意义(P<0.01);与布地奈德组比较,黄龙高剂量组血清IFN-γ值相对升高,BALF中IL-4值相对降低,IFN-γ值相对升高,差异有统计学意义(P<0.05)。结论黄龙止咳口服液通过降低机体的IL-4含量、升高IFN-γ含量发挥治疗CVA的作用。

목적:통과측정여분석각조대서혈청급폐포관세액(BALF)중γ간우소(IFN-γ)、백세포개소4(IL-4)함량,관찰황룡지해구복액대해수변이성효천(CVA)모형대서면역공능적영향,진이탐토황룡지해구복액조절CVA면역공능문란적작용궤제。방법이용란단백(OA)화경양화려공동격발치민건립CVA대서모형,장대서분위정상조、모형조、황룡고제량조、황룡중제량조、황룡저제량조、선성해천저조、포지내덕조,기록인천유해시2 min내대서해수차수급혈청화지기관BALF중IL-4、IFN-γ함량。결과여정상조비교,모형조해수차수명현증다(P<0.01),혈청화지기관BALF중IL-4함량현저승고(P<0.01),IFN-γ함량강저(P<0.01);여모형조비교,각치료조해수차수균명현감소(P<0.01),혈청급BALF중IL-4함량명현강저,IFN-γ함량명현승고,차이유통계학의의(P<0.05혹P<0.01);황룡고제량조여황룡저、중제량조비교,해수차수명현감소,혈청급BALF중IL-4함량명현강저,IFN-γ함량명현승고,차이유통계학의의(P<0.05혹P<0.01);여선성해천저조비교,황룡고제량조혈청IFN-γ치상대승고,BALF중IL-4치상대강저,IFN-γ치상대승고,차이유고도통계학의의(P<0.01);여포지내덕조비교,황룡고제량조혈청IFN-γ치상대승고,BALF중IL-4치상대강저,IFN-γ치상대승고,차이유통계학의의(P<0.05)。결론황룡지해구복액통과강저궤체적IL-4함량、승고IFN-γ함량발휘치료CVA적작용。
Objective By measuring and analyzing the content of interferon-γ (IFN-γ), interleukin-4 (IL-4) of serum and bronchoalveolar lavage fluid (BALF) in rats of all groups, to observe the effect of Huanglong Cough Liquid on im-munologic function of cough variant asthma (CVA) of model rats, so as to investigate the mechanism of Huanglong Cough Liquid in adjusting the immune dysfunction of CVA. Methods The egg albumin (OA) and aluminum hydroxide were used to motivate sensitization to establish CVA model of rats, and the rats were divided into normal group, model group, high dose of Huanglong group, medial dose of Huanglong group, low dose of Huanglong group, Xiansheng Kechuanning group, Budesonide group. The cough times and contents of IFN-γ, IL-4 of serum and bronchus BALF at inducing dyspnea with cough whin 2 min were recorded. Results Compared with normal group, the cough times of model group were increased significantly (P< 0.01), the content of IL-4 of serum and bronchus BALF was increased significantly (P<0.01), the content of IFN-γwas decreased significantly (P<0.01);compared with model group, the cough times of all groups decreased significantly (P<0.01), the content of IL-4 of serum and BALF was decreased sig-nificantly, the content of IFN-γ was increased signifi-cantly, the differences were statistically significant (P<0.05 or P < 0.01); compared with low, medial dose of Huanglong group, the cough times of high dose of Huan-glong group were decreased significantly, the content of IL-4 of serum and bronchus BALF was decreased significantly, the content of IFN-γ was increased significantly, the differences were statistically significant (P< 0.05 or P< 0.01); compared with Xiansheng Kechuanning group, the con-tent of serum IFN-γ of high dose of Huanglong group was increased relatively, the content of IL-4 of BALF was de-creased relatively, the content of IFN-γwas increased relatively, the differences were statistically significant (P<0.01);compared with Budesonide group, the content of serum IFN-γ of high dose of Huanglong group was increased relative-ly, the content of IL-4 of BALF was decreased relatively, the content of IFN-γwas increased relatively, the differences were statistically significant (P<0.05). Conclusion Huanglong Cough Liquid play a treatment role in CVA by decreas-ing the contents of IL-4 and increasing the contents of IFN-γ.
