目的:观察烟酰胺超载与氧化应激之间的关系,以确定烟酰胺在机体氧化性损伤中扮演的重要角色。方法利用长期大鼠高烟酰胺喂养模型,结合Real time PCR和Western blot方法,研究烟酰胺超载对氧化应激相关分子水平的影响。结果在烟酰胺的作用下,NNMT、DNMT1、PARP1、Catalase及P53的表达水平呈现剂量依赖性的提高;同型半胱氨酸代谢酶BHMT、CBS与MTR 出现了显著的表达补偿机制;INS1的表达受烟酰胺的影响较小。结论烟酰胺超载会触发氧化应激,这种改变可能全面并直接参与代谢综合征及其临床结局的形成和发展。

목적:관찰연선알초재여양화응격지간적관계,이학정연선알재궤체양화성손상중분연적중요각색。방법이용장기대서고연선알위양모형,결합Real time PCR화Western blot방법,연구연선알초재대양화응격상관분자수평적영향。결과재연선알적작용하,NNMT、DNMT1、PARP1、Catalase급P53적표체수평정현제량의뢰성적제고;동형반광안산대사매BHMT、CBS여MTR 출현료현저적표체보상궤제;INS1적표체수연선알적영향교소。결론연선알초재회촉발양화응격,저충개변가능전면병직접삼여대사종합정급기림상결국적형성화발전。
ObjectiveThe aim of this study was to investigate the effect of nicotinamide on the oxidative stress injury in rats.Methods Long-term high nicotinamide feeding test, together with real time PCR and Western blot were performed to investigate the effect of nicotinamide on oxidative stress injury in rats.Results Nicotinamide loading induced a significant increase in the levels of NNMT, DNMT1, PARP1, Catalase and P53; There is a compensation mechanism for homocysteine metabolism enzyme (BHMT, CBS and MTR); However, INS1 was less affected by nicotinamide.Conclusion Excess nicotinamide may trigger oxidative stress which may be directly or indirectly involved in the development of metabolic syndrome and its clinical outcomes.
