目的:探讨bag-1内含子区单核苷酸多态性(single nucleotide polymorphism, SNP)与晚期NSCLC铂类为主化疗敏感性关系。方法应用ASP-PCR技术对156例晚期NSCLC患者bag-1基因rs706120进行基因分型检测,并对其近期及远期疗效进行随访。利用SPSS软件进行数据分析。结果 bag-1 rs706120存在单核苷酸多态性;bag-1 rs706120多态性在近期疗效中差异无统计学意义(P>0.05),A/A基因型晚期NSCLC患者中位无进展生存期长于A/G及G/G基因型(163 d vs.126 d,P=0.027);与肺癌分期共同影响PFS。结论 bag-1 rs706120 SNP可能是晚期NSCLC患者铂类化疗敏感性潜在的预测因子。

Objective To investigate the relationship between SNP in bag -1intron and the sensitivity of platinum-based chemotherapy in advanced NSCLC. Methods Genotypes of rs706120 of 156 advanced NSCLC patients were classified by using ASP-PCR. The efficacy and long-term effects were recorded.Statistical analysis was performed by using SPSS. Results 1.The single nucleotide polymorphism in rs706120. 2.rs 706120 SNP showed no significant correlation with efficacy (P > 0.05). 3. rs706120 SNP reflected significant difference in the progression of free survival period, median PFS of patients with A/A genotype was longer than ones with A/G and G/G genotype (163 days vs. 126 days, P = 0.027); stage and rs706120 SNP together contributed wo the difference with PFS. Conclusion bag-1 rs706120 SNP may be a potential predictor for the sensitivity to platinum-based chemotherapy in patients with advanced NSCLC.
