目的:探讨电针对急性痛风性关节炎(AGA)大鼠受试踝关节滑膜组织髓样细胞表达激发受体(TREM)-1表达的影响。方法将40只SD雄性大鼠随机分为正常组、模型组、西药组、电针组,每组各10只;正常组常规喂养,其余采用尿酸钠溶液注射法建立AGA模型。造模前2 d,正常组与模型组按20 ml/kg予以生理盐水灌胃,西药组按1 mg/kg予以秋水仙碱溶液灌胃,电针组选取受试侧三阴交、解溪、昆仑,频率1.5~2 Hz,电压9V,电流强度1~3 mA,疏密波,留针20 min,1次/d,连续9 d。分析各组大鼠关节功能障碍,采用ELISA法检测受试踝关节滑膜组织中肿瘤坏死因子TNF-α、白细胞介素IL-1β的含量,采用免疫组化及Western blot法检测TREM-1表达。结果与正常组比较,模型组大鼠功能障碍指数显著增高(P<0.01),TNF-α和IL-1β含量显著增高(P<0.05),受试踝关节滑膜组织TREM-1表达明显增加(P<0.05);与模型组比较,西药组和电针组大鼠功能障碍指数显著降低(P<0.01),TNF-α和IL-1β含量显著降低(P<0.05),受试踝关节滑膜组织TREM-1表达明显降低(P<0.05);西药组与电针组比较,均无统计学意义(P>0.05)。结论电针治疗AGA的机制可能是通过抑制TREM-1的表达。

목적:탐토전침대급성통풍성관절염(AGA)대서수시과관절활막조직수양세포표체격발수체(TREM)-1표체적영향。방법장40지SD웅성대서수궤분위정상조、모형조、서약조、전침조,매조각10지;정상조상규위양,기여채용뇨산납용액주사법건립AGA모형。조모전2 d,정상조여모형조안20 ml/kg여이생리염수관위,서약조안1 mg/kg여이추수선감용액관위,전침조선취수시측삼음교、해계、곤륜,빈솔1.5~2 Hz,전압9V,전류강도1~3 mA,소밀파,류침20 min,1차/d,련속9 d。분석각조대서관절공능장애,채용ELISA법검측수시과관절활막조직중종류배사인자TNF-α、백세포개소IL-1β적함량,채용면역조화급Western blot법검측TREM-1표체。결과여정상조비교,모형조대서공능장애지수현저증고(P<0.01),TNF-α화IL-1β함량현저증고(P<0.05),수시과관절활막조직TREM-1표체명현증가(P<0.05);여모형조비교,서약조화전침조대서공능장애지수현저강저(P<0.01),TNF-α화IL-1β함량현저강저(P<0.05),수시과관절활막조직TREM-1표체명현강저(P<0.05);서약조여전침조비교,균무통계학의의(P>0.05)。결론전침치료AGA적궤제가능시통과억제TREM-1적표체。
Objective To investigate the effects of electro-acupuncture (EA) on the expression of triggering receptor expressed on myeloid cell(TREM)1 in ankle joint synovial tissue of acute gouty arthritis (AGA) rats. Methods Forty male SD rats were randomly divided into 4 groups:normal, AGA, medication and EA group, 10 rats in each group. AGA model was established by induced monosodium urate (MSU) method, except the normal group. Tow days before AGA model was established, normal and AGA groups were lavaged with normal saline (20 ml/kg), medication group was lavaged with colchicine solution (20 ml/kg), EA (1.5-2Hz, D.-D.wave, 9v, 1-3 mA) was applied to“Sanyinjiao”(SP6),“jiexi”(ST41) and“Kunlun”(BL60) for 20 min, once daily, continuously for 9 days. Then observed the changes in dysfunction, and the content of TNF-αand IL-1βdetected by ELISA ,the expression of TREM-1 detected by immunohistochemistry and western blot. Results Compared to the normal group, the AGA group of the dysfunction index increased significantly (P<0.01), the content of TNF-αand IL-1βincreased significantly (P<0.05), the expression of TREM-1 in synovial tissue increased significantly (P<0.05); the medication and EA groups compared to the AGA group, the dysfunction index decreased significantly (P<0.01), the content of TNF-αand IL-1βdecreased significantly (P<0.05), the expression of TREM-1 in synovial tissue decreased significantly (P<0.05); there were not statistically significant between the medication and EA group (P>0.05). Conclusion EA treating AGA may be through down-regulating the expression of TREM-1 in synovial tissue.
