目的:方法研究外源性补锌对脊髓缺血再灌注损伤大鼠 ATP酶、MAO活性和 T-AOC含量的影响,从而探讨锌对脊髓缺血再灌注损伤的保护作用的机制。方法60只 Wistar大鼠随机分为5组,每组12只,雌雄各半,分别为假手术组、模型组、硫酸锌预处理高剂量组(100 mg/kg)、中剂量组(50 mg/kg)和低剂量组(25 mg/kg)。各剂量组每日灌胃给予硫酸锌1次,连续7天。假手术组和模型组给予相同剂量生理盐水,7天后制备脊髓缺血再灌注损伤动物模型,损伤后24 h采用Basso Beattie Bresnahan(BBB)运动功能评分法对脊髓缺血再灌注损伤模型进行后肢神经功能评价;测定脊髓组织中 ATP酶、MAO活性和T-AOC含量。结果与模型组比较,各给锌组大鼠神经功能评分明显高于模型组,差异具有显著性(P<0.05)。各硫酸锌处理组 Na+-K+-ATP酶活性明显低于假手术组(P<0.05),但明显高于模型组(P<0.05)。模型组和各硫酸锌处理组脊髓组织 Ca2+-Mg2+-ATP酶活性显著低于假手术组(P<0.05)。模型组和各硫酸锌处理组大鼠脊髓组织内的 MAO活性均显著高于假手术组(P<0.05),且100 mg/kg/d 硫酸锌处理组 MAO活性显著低于模型组(P<0.05)。模型组和各硫酸锌处理组大鼠脊髓组织内的 T-AOC含量显著低于假手术组(P<0.05)。模型组大鼠脊髓组织中T-AOC含量最低,随着硫酸锌剂量的增加,T-AOC含量呈上升趋势。结论锌可能通过提高ATP酶的活性、抑制MAO活性和升高T-AOC含量,从而对脊髓缺血再灌注损伤起到一定的保护作用。

목적:방법연구외원성보자대척수결혈재관주손상대서 ATP매、MAO활성화 T-AOC함량적영향,종이탐토자대척수결혈재관주손상적보호작용적궤제。방법60지 Wistar대서수궤분위5조,매조12지,자웅각반,분별위가수술조、모형조、류산자예처리고제량조(100 mg/kg)、중제량조(50 mg/kg)화저제량조(25 mg/kg)。각제량조매일관위급여류산자1차,련속7천。가수술조화모형조급여상동제량생리염수,7천후제비척수결혈재관주손상동물모형,손상후24 h채용Basso Beattie Bresnahan(BBB)운동공능평분법대척수결혈재관주손상모형진행후지신경공능평개;측정척수조직중 ATP매、MAO활성화T-AOC함량。결과여모형조비교,각급자조대서신경공능평분명현고우모형조,차이구유현저성(P<0.05)。각류산자처리조 Na+-K+-ATP매활성명현저우가수술조(P<0.05),단명현고우모형조(P<0.05)。모형조화각류산자처리조척수조직 Ca2+-Mg2+-ATP매활성현저저우가수술조(P<0.05)。모형조화각류산자처리조대서척수조직내적 MAO활성균현저고우가수술조(P<0.05),차100 mg/kg/d 류산자처리조 MAO활성현저저우모형조(P<0.05)。모형조화각류산자처리조대서척수조직내적 T-AOC함량현저저우가수술조(P<0.05)。모형조대서척수조직중T-AOC함량최저,수착류산자제량적증가,T-AOC함량정상승추세。결론자가능통과제고ATP매적활성、억제MAO활성화승고T-AOC함량,종이대척수결혈재관주손상기도일정적보호작용。
Objective To study the effect of the exogenous zinc supplement on the ATP enzyme、MAO activity and T-AOC content of rats with spinal cord ischemia-reperfusion inj ury in order to explore the mechanism of protective effect of zinc on the spinal cord ischemia-reperfusion injury.Methods 60 Wistar rats were randomly divided into five groups:sham-operation group,model group,and zinc sulfate pre-treatment groups including high-dose zinc group (100 mg/kg),middle-dose zinc group (50 mg/kg)and low-dose zinc group(25 mg/kg),12 rats in each group,half male and half female.In all the zinc pre-treatment groups,rats were administered zinc sulfate one time by gavage in seven days succession.The rats in sham-operation group and model group were administered the same volume normal saline solu-tion as zinc pre-treatment group.Spinal cord ischemia-reperfusion inj ury model was established after zinc pretreatment for 7days.24 h after injury,Motor function assessment of rats were performed by Basso Beattie Bresnahan (BBB)Mo-tor Function Assessment;ATP enzyme,MAO activity and T-AOC content in spinal cord were detected.Results BBB motor function assessment of zinc group were significantly higher than that of model group (P<0.05).Na+-K+-ATP enzyme activity in zinc pre-treatment groups was significantly lower than that in sham-operation group(P<0.05),but significantly higher than that in model group(P<0.05).Ca2+-Mg2+-ATP enzyme activity in rats in model group and zinc pre-treatment groups was significantly lower than that in sham-operation group(P<0.05).MAO activity in rats in model group and zinc pre-treatment groups significantly higher than that in sham-operation group(P<0.05).And MAO activity in rats in high-dose zinc group (100 mg/kg)was significantly lower than that in model group (P<0.05). T-AOC activity in rats in model group and zinc pre-treatment groups was significantly lower than that in sham-opera-tion group (P<0.05),T-AOC content in model group was the lowest and increased with the increasing of zinc dose. Conclusion zinc has a protective effect on spinal cord ischemia-reperfusion inj ury by possibly.
