目的:分选口腔鳞癌干细胞并进一步研究Wnt/β-catenin信号通路在鳞癌干细胞的迁移、侵袭及eMt转化中的作用。方法磁珠分选口腔鳞癌干细胞并用细胞流式技术检测其纯度;免疫印迹检测slug、twist及oct-4的表达变化;细胞划痕及transwell小室检测口腔鳞癌干细胞的迁移及侵袭能力。结果细胞流式技术检测磁珠分选得到的cd44+细胞,结果表明分选纯度达93%;细胞划痕及transwell小室检测Wnt/β-catenin信号通路在口腔鳞癌干细胞中的作用,实验数据表明激活Wnt/β-catenin途径能够显著促进肿瘤干细胞的迁移及侵袭。免疫印迹检测发现wnt3a能够明显增强口腔鳞癌干细胞中slug、twist及oct-4的表达,同时用dKK1进行抑制实验,结果显示slug、twist及oct-4的表达降低。结论 Wnt/β-catenin信号通路在cd44+的口腔鳞癌干细胞的迁移、侵袭及eMt转化中发挥重要作用,对于肿瘤的治疗有显著的指导意义。

objective to sorting of oral squamous cell carcinoma stem cells and further research of Wnt/-catenin signaling pathway in squamous cell carcinoma invasion and migration of stem, and the role of eMt transformed cells. Magnetic beads separation Method of oral squamous cell carcinoma stem cells and detect the purity of the cell lfow cytometry;the expression change of slug, twist and Western blot detection of oct-4 and transwell assay;cell scratch migration and invasion ability of oral squamous cell carcinoma stem cells.Results cell flow detection magnetic bead sorting type technology of cd44+cells, results show that sorting purity reached 93%; cell scratch and transwell assay of Wnt/ -catenin signaling pathway in oral squamous cell carcinoma stem cells and the role, the experimental data show that the activation of Wnt/ beta -catenin pathway can significantly promote the migration and invasion of tumor stem cells. immunoblot assay showed that Wnt3a could signiifcantly enhance the expression of Slug in oral squamous cell carcinoma stem, Twist and OCT-4 cells, while DKK1 inhibition, showed decreased expression of slug, twist and oct-4.Conclusion Wnt/beta-catenin signaling pathway play an important role in invasion and migration of eMt cells, and the transformation of cd44+in oral squamous cell carcinoma stem, have signiifcant guiding signiifcance for tumor treatment.
