目的:探讨亚砷酸钠(iAs3+)染毒对家兔皮肤抗氧化水平的影响及其与砷代谢的关系。方法选取成年健康雄性家兔30只,随机分为5组,每组6只;各梁毒组家兔经口染毒,染毒剂量分别为低(L)(0.075 mg· kg-1·w-1)、中低(ML)(0.15 mg·kg-1·w-1)、中高(MH)(0.375 mg·kg-1·w-1)、高(H)(0.75 mg·kg-1· w-1),对照组(C)饮用去离子水。采用自由饮水法染毒,根据动物日饮水量配制染毒水溶液,自由饮水染毒12 w。12 w 后处死动物,取同一部位皮肤组织用2.5%戊二醛固定后进行扫描电镜观察。取家兔背部皮肤组织进行匀浆,检测谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽 S-转移酶(GST)、超氧化物歧化酶(SOD)活力。采用高效液相色谱-氢化物发生原子荧光光谱法(HPLC-HGAFS)测定尿液中砷形态代谢产物含量。结果与对照组相比,染毒组家兔体质量基本呈现增加的趋势,但差异无统计学意义(P >0.05)。电镜结果显示染毒组家兔皮肤角质层有不同程度的改变。随着 NaAsO2染毒剂量的增加,GSH-Px 活力降低,GST 活力升高,差异有统计学意义(P <0.05);SOD 差异无统计学意义(P >0.05)。尿液中 DMA、MMA 的含量随着染毒剂量的增加而升高,差异有统计学意义(P <0.05)。无机砷(iAs3+、iAs5+)与 GSH-Px、GST 无相关。GSH-Px 与 MMA 呈负相关(r =-0.507,P<0.05),与 DMA 呈负相关(r =-0.562,P <0.05)。GST 与 MMA 呈正相关(r =0.395,P <0.05),与 DMA 呈正相关(r =0.414,P <0.05)。结论砷的体内代谢过程和家兔皮肤组织的氧化与抗氧化平衡破坏有密切关系。

목적:탐토아신산납(iAs3+)염독대가토피부항양화수평적영향급기여신대사적관계。방법선취성년건강웅성가토30지,수궤분위5조,매조6지;각량독조가토경구염독,염독제량분별위저(L)(0.075 mg· kg-1·w-1)、중저(ML)(0.15 mg·kg-1·w-1)、중고(MH)(0.375 mg·kg-1·w-1)、고(H)(0.75 mg·kg-1· w-1),대조조(C)음용거리자수。채용자유음수법염독,근거동물일음수량배제염독수용액,자유음수염독12 w。12 w 후처사동물,취동일부위피부조직용2.5%무이철고정후진행소묘전경관찰。취가토배부피부조직진행균장,검측곡광감태과양화물매(GSH-Px)、곡광감태 S-전이매(GST)、초양화물기화매(SOD)활력。채용고효액상색보-경화물발생원자형광광보법(HPLC-HGAFS)측정뇨액중신형태대사산물함량。결과여대조조상비,염독조가토체질량기본정현증가적추세,단차이무통계학의의(P >0.05)。전경결과현시염독조가토피부각질층유불동정도적개변。수착 NaAsO2염독제량적증가,GSH-Px 활력강저,GST 활력승고,차이유통계학의의(P <0.05);SOD 차이무통계학의의(P >0.05)。뇨액중 DMA、MMA 적함량수착염독제량적증가이승고,차이유통계학의의(P <0.05)。무궤신(iAs3+、iAs5+)여 GSH-Px、GST 무상관。GSH-Px 여 MMA 정부상관(r =-0.507,P<0.05),여 DMA 정부상관(r =-0.562,P <0.05)。GST 여 MMA 정정상관(r =0.395,P <0.05),여 DMA 정정상관(r =0.414,P <0.05)。결론신적체내대사과정화가토피부조직적양화여항양화평형파배유밀절관계。
Objective To compare the impacts of different doses of sodium arsenite on antioxidation level in rabbit skin tissue,and to approach the correlation between the impacts and the urine arsenic metabolism level.Methods A total of 30 male healthy rabbits were divided into 5 groups:the control group (deionized water),sodium arsenite low dose group (0.075 mg/kg),secondary low dose group (0.15 mg/kg),second-ary high dose group (0.375 mg/kg),high dose group (0.75 mg/kg)respectively,with 6 rabbits in each group.Prepare the prion aqueous solution according to the rabbits′daily water intake and free drinking was exposed to 12 weeks with continuous sodium arsenite exposure.Skin tissue samples were collected in 12 weeks,and the samples were fixed with 2.5% glutaraldehyde and observed the ultrastructure by scanning electron microscope.Collect the skin tissue samples of backside and homogenate the samples.Test the ac-tivity of glutathione peroxidase (GSH-Px),glutathione transferase (GST),superoxide dismutase (SOD). High performance liquid chromatography (HPLC)-hydride generation atomic fluorescence spectrometry (HPLC-HGAFS)was adopted to test the content of arsenic metabolites in urine collected from rabbits′bladder.Results Compared with the control group,the weight of exposure groups showed an increasing tendency,but with no statistical significance (P >0.05).The results by scanning microscope showed dif-ferent changes in skin stratum corneum.The activity of GSH-Px reduced and the GST activity increased, with a statistical significance (P <0.05).There was no significance in the activity of SOD (P >0.05).The content of dimethyl arsenic acid (DMA)was higher than that of methyl arsenic acid (MMA)in urine with the sodium arsenite exposure doses increasing;the content of DMA and MMA increased first and then de-creased,with a statistical significance (P <0.05).There was no correlation between the content of inor-ganic arsenic(iAs3+ 、iAs5+ )and GSH-Px,GST respectively.There was negative correlation between the activity of GSH-Px and the content of DMA (r =-0.562,P <0.05),MMA (r =-0.507,P <0.05)re-spectively.There was positive correlation between the activity of GST and the content of DMA (r = 0.414,P<0.05),MMA (r = 0.395,P <0.05)respectively.Conclusion There proved to be close relationships between arsenic metabolism process with the oxidation and antioxidant balance destruction in rabbit skin tissues.
