Schisandra chinensis polysaccharide%isoproterenol%ventricular remodeling
目的:观察北五味子多糖(Schisandra chinensis polysaccride,SCP)抗异丙肾上腺素(Isoproterenol,ISO)诱导的心室重构作用并探讨其作用机制.方法异丙肾上腺素皮下注射致心室重构模型,经SCP低、中、高(25,50,100 mg/kg)3个剂量组和阳性对照卡托普利(50 mg/kg)治疗后,观察其对心肌病理学改变、心室重量( HW/BW)和心室重量指数( LVI)、羟脯氨酸( Hyp)含量、Ⅰ型和Ⅲ型胶原含量及Western blot技术检测心肌组织中TGF-β1表达的影响.结果 SCP不同剂量组均能降低HW/BW和LVI(P<0.05或P<0.01),降低羟脯氨酸含量(P<0.05或P<0.01)),降低心肌组织中Ⅰ型和Ⅲ型胶原含量(P<0.05或P<0.01或P<0.001),减轻TGF-β1在心肌组织中的表达(P<0.05或P<0.01或P<0.001),抑制心室重构.结论 SCP能够通过抑制TGF-β1的表达来对抗ISO诱导的大鼠心室重构.

목적:관찰북오미자다당(Schisandra chinensis polysaccride,SCP)항이병신상선소(Isoproterenol,ISO)유도적심실중구작용병탐토기작용궤제.방법이병신상선소피하주사치심실중구모형,경SCP저、중、고(25,50,100 mg/kg)3개제량조화양성대조잡탁보리(50 mg/kg)치료후,관찰기대심기병이학개변、심실중량( HW/BW)화심실중량지수( LVI)、간포안산( Hyp)함량、Ⅰ형화Ⅲ형효원함량급Western blot기술검측심기조직중TGF-β1표체적영향.결과 SCP불동제량조균능강저HW/BW화LVI(P<0.05혹P<0.01),강저간포안산함량(P<0.05혹P<0.01)),강저심기조직중Ⅰ형화Ⅲ형효원함량(P<0.05혹P<0.01혹P<0.001),감경TGF-β1재심기조직중적표체(P<0.05혹P<0.01혹P<0.001),억제심실중구.결론 SCP능구통과억제TGF-β1적표체래대항ISO유도적대서심실중구.
Objective To study the effects of Schisandra chinensis polysaccharide ( SCP ) on ventricular remodeling induced by isoproterenol ( ISO ) in rats and to explore its preliminary mechanism. Method Ventricular remodeling was induced by subcutaneous injections of ISO, myocardial pathological change was measured by HE staining, heart mass and cardiac mass index ( LVI ) of rats were calculated after the end of experiment,hydroxyproline ( Hyp ) content, and type I, III collagen contents and TGF-β1 protein expression in myocardial tissue were measured by ELISA and WB,respectively. Results Compared with those on ISO group, the heart weight index,Hyp content,collagenⅠandⅢlevels,and TGF-β1 protein expression in myocardial tissue decreased obviously(P<0. 05,P<0. 01 or P<0. 001). Conclusion Schisandra chinensis polysaccharide may restrain the ventricular remodeling induced by ISO in rats by inhibiting TGF-β1 protein expression.
