目的:原代培养猪脂肪间充质干细胞(adipose-derived mesenchymal stem cell,ADMSC),探索其适宜培养温度并对其进行诱导鉴定。方法采用胶原酶消化法原代分离培养猪脂肪间充质干细胞,用WST-1法比较猪ADMSC在37℃和38℃条件下的生长曲线,向成骨、成脂和成软骨方向诱导分化ADMSC,鉴定其多能性。结果用胶原酶消化法可以从猪颈部脂肪组织中分离培养出脂肪间充质干细胞,贴壁生长,P2代以后形态均一,稳定增殖,约5天可传代,在一定条件诱导下可以向成骨、成脂或成软骨方向分化,适宜在5%CO2、38℃、饱和湿度条件下培养。结论原代培养的猪脂肪间充质干细胞分离方便、增殖速度快、状态稳定,具有多向分化潜能,是可靠的供体细胞来源。
目的:原代培養豬脂肪間充質榦細胞(adipose-derived mesenchymal stem cell,ADMSC),探索其適宜培養溫度併對其進行誘導鑒定。方法採用膠原酶消化法原代分離培養豬脂肪間充質榦細胞,用WST-1法比較豬ADMSC在37℃和38℃條件下的生長麯線,嚮成骨、成脂和成軟骨方嚮誘導分化ADMSC,鑒定其多能性。結果用膠原酶消化法可以從豬頸部脂肪組織中分離培養齣脂肪間充質榦細胞,貼壁生長,P2代以後形態均一,穩定增殖,約5天可傳代,在一定條件誘導下可以嚮成骨、成脂或成軟骨方嚮分化,適宜在5%CO2、38℃、飽和濕度條件下培養。結論原代培養的豬脂肪間充質榦細胞分離方便、增殖速度快、狀態穩定,具有多嚮分化潛能,是可靠的供體細胞來源。
목적:원대배양저지방간충질간세포(adipose-derived mesenchymal stem cell,ADMSC),탐색기괄의배양온도병대기진행유도감정。방법채용효원매소화법원대분리배양저지방간충질간세포,용WST-1법비교저ADMSC재37℃화38℃조건하적생장곡선,향성골、성지화성연골방향유도분화ADMSC,감정기다능성。결과용효원매소화법가이종저경부지방조직중분리배양출지방간충질간세포,첩벽생장,P2대이후형태균일,은정증식,약5천가전대,재일정조건유도하가이향성골、성지혹성연골방향분화,괄의재5%CO2、38℃、포화습도조건하배양。결론원대배양적저지방간충질간세포분리방편、증식속도쾌、상태은정,구유다향분화잠능,시가고적공체세포래원。
Objective To establish a method for high yield mesenchymal stem cells collection, as well as a culture method for identifying mesenchymal stem cells from the swine adipose-derived mesenchymal stem cell (ADMSC).Methods Swine ADMSCs were isolated from fat tissue with collagenase , followed by induction of differentiation to osteogenic, adipogenic and chondrogrnic cells. The survival curve of the ADMSC at the 37 oC and 38 oC were measured using WST-1Cell Proliferation Assay Reagent.Results ADMSCs isolated with collagenase from swine neck fat tissue generated a stable uniform appearance after the second generation. The passage period was ifve days. ADMSC could differentiate into osteogenic, adipogenic or chondrogrnic cells under different culture conditions. The highest growth rate was achieved at 38 oC in this study.Conclusion Swine ADMSCs have the potential to differentiate into osteogenic, adipogenic or chondrogrnic cells, and they may be appropriate for transplantation for both research and clinical purpose.
