目的:研究不同剂量氨氯地平抑制小鼠黑色素瘤高转移细胞株B16转移的作用,并探讨其作用机制。方法:选取64只C57BL/6J小鼠,将黑色素瘤髙转移细胞株B16接种在小鼠腹股沟皮下(2×106个/只)。将接种后的小鼠以随机数字表法分为对照组和氨氯地平高、中、低剂量治疗组,对照组小鼠仅给予0.9%氯化钠注射液;治疗组小鼠给予不同剂量的氨氯地平治疗,分别为1 mg/kg(低剂量组)、3 mg/kg(中剂量组)和10 mg/kg(高剂量组)。观察各组小鼠肿瘤生长情况及身体情况;观察各组小鼠肿瘤肺转移情况,计算肺转移的结节及肿瘤抑瘤率;采用免疫组织化学法检测各组小鼠肿瘤组织中白细胞介素8(IL-8)蛋白表达水平;采用逆转录-聚合酶链反应(RT-PCR)检测各组小鼠肿瘤组织中IL-8的信使核糖核酸(mRNA)表达水平。结果:氨氯地平对小鼠黑色素瘤高转移细胞株B16的自发性肺转移有显著的抑制作用;治疗组小鼠肿瘤组织中IL-8蛋白表达均明显减弱,且随氨氯地平剂量的增加而减弱;治疗组小鼠肿瘤组织中IL-8的mRNA的表达明显低于对照组,且IL-8的mRNA的表达随氨氯地平剂量的增加而减弱。结论:氨氯地平对小鼠黑色素瘤高转移细胞株B16有一定的抑瘤和抑制肺转移作用,其作用可能与影响IL-8的表达有关。

목적:연구불동제량안록지평억제소서흑색소류고전이세포주B16전이적작용,병탐토기작용궤제。방법:선취64지C57BL/6J소서,장흑색소류고전이세포주B16접충재소서복고구피하(2×106개/지)。장접충후적소서이수궤수자표법분위대조조화안록지평고、중、저제량치료조,대조조소서부급여0.9%록화납주사액;치료조소서급여불동제량적안록지평치료,분별위1 mg/kg(저제량조)、3 mg/kg(중제량조)화10 mg/kg(고제량조)。관찰각조소서종류생장정황급신체정황;관찰각조소서종류폐전이정황,계산폐전이적결절급종류억류솔;채용면역조직화학법검측각조소서종류조직중백세포개소8(IL-8)단백표체수평;채용역전록-취합매련반응(RT-PCR)검측각조소서종류조직중IL-8적신사핵당핵산(mRNA)표체수평。결과:안록지평대소서흑색소류고전이세포주B16적자발성폐전이유현저적억제작용;치료조소서종류조직중IL-8단백표체균명현감약,차수안록지평제량적증가이감약;치료조소서종류조직중IL-8적mRNA적표체명현저우대조조,차IL-8적mRNA적표체수안록지평제량적증가이감약。결론:안록지평대소서흑색소류고전이세포주B16유일정적억류화억제폐전이작용,기작용가능여영향IL-8적표체유관。
OBJECTIVE:To study the mechanism underlying the inhibitory effect of amlodipine on melanoma cell line B16.METHODS: The groin region in a total of 64 C57BL/6J mice was inoculated subcutaneously with highly metastatic melanoma cell line B16 (2 ×106 cells/rats) then the mice were randomized to receive either 0.9%sodium chloride injection(control group) or amlodipine [1 mg/kg(low dose group), 3 mg/kg(medium dose group) or 10 mg/kg ( high dose group ) ] .The tumor growth and physical conditions of mice were followed; pulmonary metastases was followed with nodules in pulmonary metastasis and tumor inhibition rate calculated ;immunohistochemistry was employed to detect the level of interleukin-8(IL-8) protein in tumor tissue, and reverse transcription-polymerase chain reaction (RT-PCR) was employed to detect the level of mRNA in IL-8 in tumor tissue.RESULTS: Amlodipine exhibited significant inhibitory effect on the spontaneous pulmonary metastases of the highly metastatic melanoma cell line B 16 in mice;the expression of IL-8 protein in the treatment group significantly decreased in dose-dependent manner; mRNA expression of IL-8 was significantly lower in the treatment groups than in the control group , and the mRNA expression of IL-8 decreased with the increase of the doses of amlodipine .CONCLUSIONS: The inhibitory effect of amlodipine on highly metastatic melanoma cell line B 16 in mice might be associated with its impact on the expression of IL-8.
