目的:优化甲醛对灭活丙型副伤寒沙门菌原液的条件,确定灭活工艺参数。方法在37℃条件下,比较不同甲醛浓度、灭活时间对丙型副伤寒沙门菌原液的灭活效果,依据无菌检查、外观以及效价测定的结果,确定丙型副伤寒沙门菌原液的灭活工艺参数,并对该工艺进行细菌灭活验证。结果不同菌株制成的丙型副伤寒沙门菌液,当稀释至一定浓度(<1000×108 cfu/mL)后,采用终体积分数为1%的甲醛溶液,经37℃灭活48 h,可达到完全灭菌的效果,同时维持了抗原的稳定性。结论经过条件优化,确定了丙型副伤寒沙门菌灭活的工艺参数,完善了关键技术。

목적:우화갑철대멸활병형부상한사문균원액적조건,학정멸활공예삼수。방법재37℃조건하,비교불동갑철농도、멸활시간대병형부상한사문균원액적멸활효과,의거무균검사、외관이급효개측정적결과,학정병형부상한사문균원액적멸활공예삼수,병대해공예진행세균멸활험증。결과불동균주제성적병형부상한사문균액,당희석지일정농도(<1000×108 cfu/mL)후,채용종체적분수위1%적갑철용액,경37℃멸활48 h,가체도완전멸균적효과,동시유지료항원적은정성。결론경과조건우화,학정료병형부상한사문균멸활적공예삼수,완선료관건기술。
Objective To optimize the conditions in inactivation of salmonella paratyphoid C bulk with formaldehyde solu-tion,and determine the parameters of the inactivation process.Methods The inactivation effects of salmonella paraty-phoid C bulk from different concentration of formaldehyde solution and different inactivation time were compared at 37 ℃, parameters of the inactivation process were determined according to the results of appearance,sterility and titer test,and the validation for bacterial inactivation was carried out.Results Salmonella paratyphoid C bulk diluted to a certain concentration(<1 000×108 cfu/mL),inactivated by formaldehyde solution at a final concentration of 1% (V /V),and in-cubated at 37 ℃ for 48 h,the bulk could be achieved a complete sterilization,and preserved antigeicity in stable. Conclusion After optimization,the parameters in inactivation process of salmonella paratyphoid C were determined,and the key technique was improved.
