采用聚丙烯酰胺凝胶电泳技术,分析了兰州鲇(Silurus lanzhouensis)肌肉、心脏、肝脏、脑、脾脏、肾脏和眼睛等7种组织中乳酸脱氢酶(LDH)、酯酶(EST)、苹果酸脱氢酶(MDH)、醇脱氢酶(ADH)、山梨醇脱氢酶(SDH)和谷氨酸脱氢酶(GDH)等6种同工酶的表达模式,并对各同工酶的酶谱进行了分析。结果表明:这6种同工酶在兰州鲇7种组织中的分布均存在明显的组织特异性;6种酶共记录出17个基因位点,其中 EST-2、 EST-3、 MDH-3和 SDH-2为多态位点;兰州鲇群体的多态位点比例 P 为23.53%,位点平均有效等位基因数 Ne 为1.24;平均观测杂合度和平均期望杂合度分别为0.1372和0.1051, Hardy-Weinberg 遗传偏离指数D 为0.3054,杂合子处于过剩状态。从以上数据中可以看出,兰州鲇野生群体遗传变异程度较大,但是有效等位基因数偏低,有必要对兰州鲇野生种质资源及时加以保护。

Polyacrylamide gel electrophoresis was used to investigate the expression pattern and isozyme zymogram of six isozymes (LDH, EST, MDH, ADH, SDH and GDH) in 7 tissues (muscle, heart, liver, brain, spleen, kidney and eye) of Silurus lanzhouensis.The results showed that: the expression of all the isozymes studied were highly tissue -specific in S.lanzhouensis; 17 enzyme-coding loci were examined, of which four loci were polymorphic , i.e., EST-2, EST-3, MDH-3 and SDH-2; the proportion of polymorphic loci was 23.53%, while the average effective number of alleles (Ne) was1.24; the average values of observed and expected heterozygosity were 0.137 2 and 0.105 1, respectively; the Har-dy-Weinberg deviation index was 0.305 4, which showed that heterozygotes were surplus .The above data indicated that the genetic variation ofS.lan zhouensis wild population was relatively high ; however, the average effective number of alleles was quite low.The protection of wild resources of S.lanzhouensis should be conducted in time.
