为了揭示高 pH 对河蟹体色的潜在影响,探讨了高 pH 胁迫下河蟹虾青素和抗氧化能力的变化。将河蟹饲养于6个规格为18 m 2的水泥池中,养殖用水为沙滤河水(pH 7.5~8.0)。将其中三个水泥池的水用1 mol/L NaOH 调 pH 至9.0~9.5。分别于 pH 调整后的2、24、48、96、192 h 时采集河蟹血淋巴和头胸甲,分析头胸甲中虾青素含量、血清超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)活性和总抗氧化力(T-AOC)。结果表明:pH 胁迫后96 h 内河蟹头胸甲中虾青素含量显著下降(P <0.05),96 h 后虾青素含量趋于稳定;血清 SOD、 CAT、T-AOC 呈现先上升后下降的趋势, SOD 和 CAT 均在胁迫48 h 后与对照组差异不显著, T-AOC 在胁迫192 h 时与对照组差异不显著。实验结果表明高 pH 胁迫会导致河蟹氧化应激,从而使河蟹体色变差。

The lush aquatic plants grown in aquaculture ponds of Eriocheir sinensis cause high pH of pond water.In order to reveal the potential impact of the high pH on crab color , this study explored the response of crab astaxanthin content and antioxidant capacity to high pH.Crabs were reared in six cement pools(18 m 2 in size), and river water (pH7.5 ~8.0) was used after filtering by sand.The water pH of three cement pools was modulated to 9.0 to 9.5using 1 mol/L NaOH. Hemolymph and carapace of crabs were sampled at 2, 24, 48, 96, 192 h respectively after pH modulation.Astaxanthin content of carapace and the activity of serum superoxide dismutase (SOD), catalase (CAT) activity and total antioxidant capacity (T-AOC) were determined.The results showed that pH stress made the astaxanthin content of crab carapace de -crease significantly during 96 h (P <0.05), however, no further significant decrease was observed after 96 h.The SOD, CAT and T-AOC activity of serum present the trend of rising firstly and declining afterward .No significant decrease was ob-served in SOD and CAT after 48 h of stress, and no significant difference in T-AOC was detected after 192 h of stress com-pare with control group (P >0.05).These results indicated that high pH contributed poor color by arousing oxidative stress of crabs.
