目的:探讨风湿性心脏瓣膜病合并心房颤动( AF)患者DNA甲基转移酶3A(DNMT3A)与超速延迟整流性钾通道蛋白(Kv1.5)表达水平变化及其相关性研究。方法将20例风湿性心脏瓣膜病接受瓣膜置换手术患者于体外循环前切取的右心耳组织作为研究对象,根据AF的定义将标本分为AF组(n=10)和窦性心律(SR)组(n=10),分别应用免疫组化、Western blot 法检测心房肌组织中 DNMT3A 和Kv1.5蛋白表达水平变化。结果免疫组化检测显示AF组与SR组相比,DNMT3A蛋白表达增加(P<0.05);而Kv1.5蛋白表达减少(P<0.05)。同时,Western blot检测显示AF组与 SR 组相比,DNMT3A 蛋白表达增加(P <0.05),而Kv1.5蛋白表达减少(P<0.05)。相关性分析显示DNMT3A与Kv1.5之间存在一定负相关性(r =-0.64, P <0.01)。结论 DNMT3A与Kv1.5之间存在负相关性,提示DNMT3A可能参与调控Kv1.5通道蛋白的表达。

목적:탐토풍습성심장판막병합병심방전동( AF)환자DNA갑기전이매3A(DNMT3A)여초속연지정류성갑통도단백(Kv1.5)표체수평변화급기상관성연구。방법장20례풍습성심장판막병접수판막치환수술환자우체외순배전절취적우심이조직작위연구대상,근거AF적정의장표본분위AF조(n=10)화두성심률(SR)조(n=10),분별응용면역조화、Western blot 법검측심방기조직중 DNMT3A 화Kv1.5단백표체수평변화。결과면역조화검측현시AF조여SR조상비,DNMT3A단백표체증가(P<0.05);이Kv1.5단백표체감소(P<0.05)。동시,Western blot검측현시AF조여 SR 조상비,DNMT3A 단백표체증가(P <0.05),이Kv1.5단백표체감소(P<0.05)。상관성분석현시DNMT3A여Kv1.5지간존재일정부상관성(r =-0.64, P <0.01)。결론 DNMT3A여Kv1.5지간존재부상관성,제시DNMT3A가능삼여조공Kv1.5통도단백적표체。
Objective To investigate the expression correlation between DNA methyltransferase 3 A ( DNMT3 A ) and rapid delayed rectifier potassium channel(Kv1. 5) in the patients with rheumatic heart disease and atrial fibril-lation ( AF) . Methods Tissues of auricula dextra were removed from 20 patients with rheumatic heart disease un-dergoing cardiac surgery for correction to valvular defect. According to the definition of AF ( n =10 ) ,20 patients were divided into groups of AF and of sinus rhythm(SR)(n=10). Expressions of DNMT3A and Kv1. 5 were de-tected by immunohistochemistry and Western blot. Results Compared with the group of SR, the expression of DN-MT3A protein was significantly enhanced (P<0. 05) whereas Kv1. 5 decreased(P<0. 05) in the results by immu-nohistochemistry. Meanwhile, the results from Western blot also showed increased DNMT3A(P <0. 05) and re-duced Kv1. 5(P<0. 05) in the group of AF. And negative correlation between the expression of DNMT3A and Kv1. 5 was shown with correlation analysis(r= -0. 64, P<0. 01). Conclusion The negative correlation exists in the expression of DNMT3 A and Kv1 . 5 , which indicates that DNMT3 A can regulate the expression of Kv1 . 5 .
