为了筛选出适应于口蹄疫疫苗生产的最佳细胞株,本研究用自制的 GM EM 培养基,以30 mL/L(v/v)血清浓度,在同等条件下比较了3种不同来源的BHK21悬浮细胞培养特性。结果显示,这3种细胞在形态上无明显的差异,在显微镜下不易区分;在培养过程中,BHK21‐L细胞株在最短时间内可达到最大培养密度,细胞生长速度快,细胞活性较高,DNA合成旺盛;BHK21C13‐2P细胞株最大密度较低,在72 h~96 h被完全阻止在G0/G1,放大比例较小,为1∶1.5~1∶2;BHK21‐B细胞在72 h~96 h停止增殖,可放大较大比例,但易出现细胞结团、易贴壁的现象,影响细胞的增殖。可见,B H K21‐L细胞是生产口蹄疫疫苗可供选择的最佳细胞株。

In order to select the best cell line adapted to the productionof FMD vaccine ,the research com‐pared the culture characteristics of three kinds of different strains from BHK‐21 suspension cells under the same conditions with GMEM culture medium ,30 mL/L serum concentration .The results showed that ,no obvious difference between these three kinds of cells in morphology ,the cells can not be easily distinguish‐ed under the microscope .In the process of cultivation ,the BHK21‐L cell line can reach the maximum den‐sity in the shortest time ,the growth rate and cell activity is high ,DNA synthesis is exuberant .The maxi‐mum density of BHK21C13‐2P cell line is lower ,in the 72 h‐96 h was completely blocked in G0/G1 ,ampli‐fying smaller proportion ,1∶1 .5‐1∶2 ;BHK21‐B cells stopped proliferation in 72 h‐96 h ,can enlarge a larger proportion ,but be prone to cell cluster ,easy adherent phenomenon ,affect cell growth .Therefore , BHK21‐L cell is the best cell line producing cells for foot‐and‐mouth disease vaccine .
