为筛选奶山羊乳房炎灭活疫苗的有效佐剂,本研究以大肠埃希菌、金黄色葡萄球菌、凝固酶阴性表皮葡萄球菌及产色葡萄球菌为材料,经甲醛37℃灭活,分别与铝胶盐、蜂胶和转移因子混合,制备铝胶盐佐剂疫苗、蜂胶佐剂疫苗和转移因子佐剂疫苗,免疫泌乳期奶山羊,免疫剂量为2 mL ,共免疫2次,免疫间隔14 d。分别在免疫前及每次免疫后第10天采集血样和乳样,ELISA 测定抗体效价,用快速诊断液检测隐性乳房炎,统计隐性乳房炎检出率。结果显示,铝胶盐佐剂疫苗刺激机体产生的抗体水平在首免和二免后均显著高于对照组(P<0.05);3个免疫组中,铝胶盐组抗体水平最高,转移因子组次之,蜂胶组则不显著;铝胶盐佐剂疫苗还能显著降低隐性乳房炎的检出率,使隐性乳房炎检出率由免疫前的60%下降至二免后的30%。结果表明,同蜂胶和转移因子相比,铝胶盐佐剂能刺激机体产生更强的体液免疫应答,对隐性乳房炎的治疗效果更显著。

위사선내산양유방염멸활역묘적유효좌제,본연구이대장애희균、금황색포도구균、응고매음성표피포도구균급산색포도구균위재료,경갑철37℃멸활,분별여려효염、봉효화전이인자혼합,제비려효염좌제역묘、봉효좌제역묘화전이인자좌제역묘,면역비유기내산양,면역제량위2 mL ,공면역2차,면역간격14 d。분별재면역전급매차면역후제10천채집혈양화유양,ELISA 측정항체효개,용쾌속진단액검측은성유방염,통계은성유방염검출솔。결과현시,려효염좌제역묘자격궤체산생적항체수평재수면화이면후균현저고우대조조(P<0.05);3개면역조중,려효염조항체수평최고,전이인자조차지,봉효조칙불현저;려효염좌제역묘환능현저강저은성유방염적검출솔,사은성유방염검출솔유면역전적60%하강지이면후적30%。결과표명,동봉효화전이인자상비,려효염좌제능자격궤체산생경강적체액면역응답,대은성유방염적치료효과경현저。
In order to select an effective adjuvant for inactivated vaccine against dairy goat mastitis ,E .coli , Staphylococcus aureus ,coagulase negative Staphylococcus epidermidis and Staphylococcus chromogenes were inactivated by using methanal at 37 ℃ and then thoroughly mixed with aluminium salt ,propolis or transfer factor to prepare aluminium salt ,propolis or transfer factor adjuvant vaccines .Each lactating dair‐y goat was inoculated subcutaneously via neck twice with a interval of 14 d and the dosage was 2 mL each time .Blood and milk samples were collected before and 10 days after each inoculation .The levels of anti‐bodies were measured by an indirect ELISA while the numbers of goats suffering subclinical mastitis were measured by a rapid diagnostic reagent .The results showed that antibodies against E .coli ,Staphylococcus aureus ,coagulase negative Staphylococcus epidermidis and Staphylococcus chromogenes in blood and milk of aluminium group were significantly higher than that of control group (P<0 .05) ,and aluminum group's the antibody levels were highest with the transfer factor group and propolis group followed .Aluminum ad‐juvant vaccine can significantly reduce the detection rate of subclinical mastitis ,and the rate was dropped from 60% before the immunization to 30% after the second injection .In conclusion ,compared with propo‐lis and transfer factor ,aluminum adjuvant can stimulate the body to produce stronger humoral immune re‐sponse and has a significant therapeutic effect to subclinical mastitis .
