目的:探讨应用缬沙坦对糖尿病小鼠肾小球组织中 Notch通路活化及足细胞丢失的影响。方法建立糖尿病小鼠模型给予缬沙坦治疗,观察小鼠肾小球细胞凋亡及足细胞脱落情况,采用Western blot检测Notch胞内域1(Notch intracellular domain 1,NICD1)、Hes1、Hey1、Bax、Bcl-2、cleaved caspase-3蛋白表达情况,Real-time PCR检测 Hes1和 Hey1 mRNA表达情况。结果缬沙坦可降低糖尿病小鼠肾小球细胞凋亡及足细胞脱落,减少糖尿病小鼠肾小球组织中 NICD1、Hes1、Hey1、Bax、cleaved caspase-3的表达,增加Bcl-2表达(P<0.05)。结论缬沙坦可通过抑制糖尿病小鼠肾小球组织中 Notch通路的活化,减少足细胞凋亡和脱落。

목적:탐토응용힐사탄대당뇨병소서신소구조직중 Notch통로활화급족세포주실적영향。방법건립당뇨병소서모형급여힐사탄치료,관찰소서신소구세포조망급족세포탈락정황,채용Western blot검측Notch포내역1(Notch intracellular domain 1,NICD1)、Hes1、Hey1、Bax、Bcl-2、cleaved caspase-3단백표체정황,Real-time PCR검측 Hes1화 Hey1 mRNA표체정황。결과힐사탄가강저당뇨병소서신소구세포조망급족세포탈락,감소당뇨병소서신소구조직중 NICD1、Hes1、Hey1、Bax、cleaved caspase-3적표체,증가Bcl-2표체(P<0.05)。결론힐사탄가통과억제당뇨병소서신소구조직중 Notch통로적활화,감소족세포조망화탈락。
Objective To investigate the effect of valsartan on activation of Notch pathway and podocyte loss.Methods Apoptosis and podocyte detachment of glomeruli were measured after diabetic mice were treated with valsartan.The levels of Notch intracellular domain 1 (NICD1),Hes1,Hey1,Bax,Bcl-2 and cleaved caspase-3 were determined by Western blot.Real-time PCR was used to evaluate Hes1 and Hey1 mRNA expression.Results Valsartan suppressed cell apoptosis and podocyte detachment of glomeruli in diabetic mice.Valsartan also inhibited overexpression of NICD1,Hes1,Hey1,Bax and cleaved caspase-3,and increased Bcl-2 level in glomerular tissues of diabetic mice (P<0.05).Conclusion Valsartan inhibits the activation of Notch pathway in glomerular tissues of diabetic mice,also inhibits podocyte apoptosis and detachment.
