目的:建立一种简单、快速的同步测定氟西汀和氯氮平血药浓度的高效液相色谱方法。方法:以氯丙咪嗪为内标,样品用含2%异丙醇的正庚烷提取净化,以C18柱作为分析柱,流动相为甲醇-磷酸氢二铵缓冲液(70∶30), pH 6.5,流速为1.0 mL/min ,柱温为40℃,检测波长为230 nm。结果:氟西汀和氯氮平的线性范围为50~1500μg/L ,线性关系良好,分析方法的检测限为10.0μg/L。低、中、高浓度的回收率、稳定性及精密度均符合方法学要求。结论:该方法精密、简便、快速,可用于临床血药浓度监测和药代动力学研究。

목적:건립일충간단、쾌속적동보측정불서정화록담평혈약농도적고효액상색보방법。방법:이록병미진위내표,양품용함2%이병순적정경완제취정화,이C18주작위분석주,류동상위갑순-린산경이안완충액(70∶30), pH 6.5,류속위1.0 mL/min ,주온위40℃,검측파장위230 nm。결과:불서정화록담평적선성범위위50~1500μg/L ,선성관계량호,분석방법적검측한위10.0μg/L。저、중、고농도적회수솔、은정성급정밀도균부합방법학요구。결론:해방법정밀、간편、쾌속,가용우림상혈약농도감측화약대동역학연구。
Objective: To develop a simple and rapid method for synchronous determination of serum drug concentration of fluoxetine and clozapine in psychiatric patient by HPLC . Methods: Chloride imipramine was used as a internal standard , serum samples were extracted by n-heptane containing 2% isopropyl alcohol , the C18 analytical column was adopted with the mobile phase of methanol sodium phosphate dibasic buffer solution (70∶30), the pH was 6 . 5 , the flow rate was 1 . 0 mL/min , the column temperature was at 40 ℃ and the detective UV wavelength was at 230 nm . Results: The linearity was obtained within the range of 50 ~ 1 500 μg/L of both fluoxetine and clozapine , which was good in linear relationship . The minimal quantification limit was 10 . 0 μg/L . The recovery , stability and precision of serum drug concentrations at low , middle and high levels were all in line with the requirements of the methodology . Conclusion: This method is simple , precise and rapid , it can be used for monitoring serum drug concentration in clinical practice and pharmacokinetic study .
