Paris polyphylla var. yunnanensis%fatty acids%nasopharyngeal carcinoma%antiproliferative activities
目的:研究云南重楼乙醇提取物的化学成分及其抑制鼻咽癌细胞的活性。方法采用反复硅胶柱层析和Sephadex LH-20等色谱方法对云南重楼的大孔树脂60%(体积分数)乙醇洗脱部位进行分离、纯化,通过理化常数和波谱分析鉴定化合物结构;采用MTT比色法进行体外抑制鼻咽癌细胞( CNE)活性测试。结果从云南重楼的大孔树脂60%(体积分数)乙醇洗脱部位分离鉴定出2个脂肪酸类化合物,分别为(8R?,9R?,10S?,6Z)-trihydroxyoctadec-6-enoic acid(1)和methyl (9S,10R,11S)-trihydroxy-12(Z)-octadecenoate(2);活性测试结果显示,化合物2具有一定的抑制鼻咽癌细胞活性,其IC50为20.77μmol ·L-1。结论2个脂肪酸均为首次从该属植物中分离得到。

Objective To study on the chemical constituents of the alcoholic extract of Paris polyphylla var. yunnanensis and their antiproliferative activities on nasopharyngeal carcinoma epithelia cell ( CNE ) . Methods The constituents were isolated and purified by repeated column chromatographies including silica gel and sephadex LH-20. Their structures were elucidated by their physicochemical properties and spectral data. The identified compounds were screened for their antiproliferative activities on CNE by using the 3-( 4,5-dimethylthiazol-2-yl )-2, 5-diphenyl tetrazolium bromide ( MTT ) assay. Results Two hydroxy fatty acids were obtained from the 60% ethanol eluting fraction of macroporous resin, ( 8R?,9R?,10S?,6Z)-trihydroxyoctadec-6-enoic acid (1)and methyl (9S,10R,11S)-trihydroxy-12(Z)-octadecenoate (2). The antiproliferative test results indicated that compound 2 showed weak antiproliferative effect on CNE cells with IC50 value of 20.77μmol·L-1 . Conclusion Compounds 1-2 were isolated from the genus Paris for the first time.
