目的:考察四妙勇安汤中绿原酸在大鼠肠的吸收动力学特征。方法采用大鼠在体肠灌流模型,结合高效液相色谱法和紫外分光光度法分别测定绿原酸和酚红的质量浓度。结果绿原酸在十二指肠、回肠、空肠的吸收速率常数分别为0.0816、0.0912、0.0748 h-1,回肠与十二指肠、空肠比较,差异均有统计学意义(P<0.01);绿原酸吸收动力学方程为Y=-0.0886X+2.8993(r=0.9932),吸收速率常数为0.0886 h-1。结论绿原酸在回肠段的吸收优于十二指肠段和空肠段,药物吸收呈一级动力学过程。
目的:攷察四妙勇安湯中綠原痠在大鼠腸的吸收動力學特徵。方法採用大鼠在體腸灌流模型,結閤高效液相色譜法和紫外分光光度法分彆測定綠原痠和酚紅的質量濃度。結果綠原痠在十二指腸、迴腸、空腸的吸收速率常數分彆為0.0816、0.0912、0.0748 h-1,迴腸與十二指腸、空腸比較,差異均有統計學意義(P<0.01);綠原痠吸收動力學方程為Y=-0.0886X+2.8993(r=0.9932),吸收速率常數為0.0886 h-1。結論綠原痠在迴腸段的吸收優于十二指腸段和空腸段,藥物吸收呈一級動力學過程。
목적:고찰사묘용안탕중록원산재대서장적흡수동역학특정。방법채용대서재체장관류모형,결합고효액상색보법화자외분광광도법분별측정록원산화분홍적질량농도。결과록원산재십이지장、회장、공장적흡수속솔상수분별위0.0816、0.0912、0.0748 h-1,회장여십이지장、공장비교,차이균유통계학의의(P<0.01);록원산흡수동역학방정위Y=-0.0886X+2.8993(r=0.9932),흡수속솔상수위0.0886 h-1。결론록원산재회장단적흡수우우십이지장단화공장단,약물흡수정일급동역학과정。
Objective To study the intestinal absorption kinetics of chlorogenic acid in Simiao Yongan decoction in rats. Methods The study was performed by an in vivo intestinal perfusion model in rats, and the concentration of chlorogenic acid and phenol red were determined by high performance liquid chromatography and ultraviolet spectroscopy, respectively. Results The intestinal absorption rate constants at duodenum,ileum and jejunum were 0.081 6, 0.091 2, 0.074 8 h-1 , and there was significant difference between duodenum,ileum and jejunum on the absorption of chlorogenic acid ( P<0.01) . The regression of chlorogenic acid was Y=-0.088 6X+2.899 3(r=0.993 2),and the absorption rate was 0. 088 6 h-1. Conclusion The absorption of chlorogenic acid at ileum is superior to that at duodenum and jejunum,and the absorption of chlorogenic acid is a first-order kinetics process.
