目的:研究金雀异黄素能否和如何抑制肝细胞癌MHCC97H细胞系肝癌干细胞样细胞侵袭。方法:干细胞条件培养基悬浮培养MHCC97 H细胞系球形成细胞,作为肝癌干细胞样细胞。不同浓度金雀异黄素处理后,Transwell小室侵袭试验检测体外细胞侵袭能力;Western blot分析Gli1和Snail1蛋白表达。结果:干细胞条件培养基悬浮培养MHCC97H细胞能形成肿瘤球。金雀异黄素(5、10和20μM)作用第3代球形成细胞即肝癌干细胞样细胞72 h降低肿瘤球形成率和细胞侵袭率;呈浓度依赖性。金雀异黄素(5、10和20μM)孵育肝癌干细胞样细胞24 h下调Gli1和Snail1蛋白表达。结论:金雀异黄素可能通过调控Gli1抑制Snail1蛋白表达抑制肝癌干细胞样细胞侵袭。
目的:研究金雀異黃素能否和如何抑製肝細胞癌MHCC97H細胞繫肝癌榦細胞樣細胞侵襲。方法:榦細胞條件培養基懸浮培養MHCC97 H細胞繫毬形成細胞,作為肝癌榦細胞樣細胞。不同濃度金雀異黃素處理後,Transwell小室侵襲試驗檢測體外細胞侵襲能力;Western blot分析Gli1和Snail1蛋白錶達。結果:榦細胞條件培養基懸浮培養MHCC97H細胞能形成腫瘤毬。金雀異黃素(5、10和20μM)作用第3代毬形成細胞即肝癌榦細胞樣細胞72 h降低腫瘤毬形成率和細胞侵襲率;呈濃度依賴性。金雀異黃素(5、10和20μM)孵育肝癌榦細胞樣細胞24 h下調Gli1和Snail1蛋白錶達。結論:金雀異黃素可能通過調控Gli1抑製Snail1蛋白錶達抑製肝癌榦細胞樣細胞侵襲。
목적:연구금작이황소능부화여하억제간세포암MHCC97H세포계간암간세포양세포침습。방법:간세포조건배양기현부배양MHCC97 H세포계구형성세포,작위간암간세포양세포。불동농도금작이황소처리후,Transwell소실침습시험검측체외세포침습능력;Western blot분석Gli1화Snail1단백표체。결과:간세포조건배양기현부배양MHCC97H세포능형성종류구。금작이황소(5、10화20μM)작용제3대구형성세포즉간암간세포양세포72 h강저종류구형성솔화세포침습솔;정농도의뢰성。금작이황소(5、10화20μM)부육간암간세포양세포24 h하조Gli1화Snail1단백표체。결론:금작이황소가능통과조공Gli1억제Snail1단백표체억제간암간세포양세포침습。
Objective To investigate whether and how genistein suppresses the invasion of hepatic cancer stem-like cells(HCSLCs) from MHCC97H cell line.Methods Sphere-forming cells(SFCs) derived from MH-CC97 H cell line which is also called HCSLCs were obtained though suspended culture with stem cell -conditioned medium.After treatment with various concentrations of genistein , the capability of invasion was tested using Tran-swell invasion assay .And the protein expression of Gli 1 and Snail1 was analyzed by western blot .Results MH-CC97 H cells can form tumor spheres though suspended culture with stem cell-conditioned medium .After treatment with genistein(5, 10 and 20 μM) for 72h, the rate of sphere-forming and invasion was decreased in SFCs namely HCSLCs, in a concentration-dependent manner .And the protein expression of Gli1 and Snail1 was down-regulated in HCSLCs.Conclusion Effects of genistein on inhibiting invasion of HCSLCs is likely involved in the reduced protein expression of Snail1 by regulating Gli1.
