目的:合成氢化荧光素二乙酸酯,并用于肿瘤细胞的染色标记。方法:通过锌粉和冰醋酸还原荧光素制备二氢荧光素,再经乙酸酐酰化合成氢化荧光素二乙酸酯。氢化荧光素二乙酸酯对肿瘤细胞进行染色,并与荧光素二乙酸酯的染色结果进行对照。结果:不论是贴壁还是悬浮细胞,当浓度为1.0 mg/L孵育染色5 min时,FHDA可基本标记细胞,背景低,可定量检测细胞的荧光强度。结论:氢化荧光素二乙酸酯可用于肿瘤细胞的荧光标记及活性氧等细胞活性物质的检测。
目的:閤成氫化熒光素二乙痠酯,併用于腫瘤細胞的染色標記。方法:通過鋅粉和冰醋痠還原熒光素製備二氫熒光素,再經乙痠酐酰化閤成氫化熒光素二乙痠酯。氫化熒光素二乙痠酯對腫瘤細胞進行染色,併與熒光素二乙痠酯的染色結果進行對照。結果:不論是貼壁還是懸浮細胞,噹濃度為1.0 mg/L孵育染色5 min時,FHDA可基本標記細胞,揹景低,可定量檢測細胞的熒光彊度。結論:氫化熒光素二乙痠酯可用于腫瘤細胞的熒光標記及活性氧等細胞活性物質的檢測。
목적:합성경화형광소이을산지,병용우종류세포적염색표기。방법:통과자분화빙작산환원형광소제비이경형광소,재경을산항선화합성경화형광소이을산지。경화형광소이을산지대종류세포진행염색,병여형광소이을산지적염색결과진행대조。결과:불론시첩벽환시현부세포,당농도위1.0 mg/L부육염색5 min시,FHDA가기본표기세포,배경저,가정량검측세포적형광강도。결론:경화형광소이을산지가용우종류세포적형광표기급활성양등세포활성물질적검측。
Objective Synthesis of fluorescindiacetate and staining cancer cells .Methods Fluorescein was reduced to fluorescin by zinc powder and ice acetic acid , fluorescindiacetate was prepared by acetation of fluoresce-in with acetic anhydride .Cancer cells were stained with fluorescindiacetate and campared with the results that stained with fluoresceindiacetate .Results Adherent cells and suspension cells could be all stain when incubating and staining 5 mins with 1.0 mg/Lfluorescindiacetate.Fluorescent intensity in cells could be observed because of low fluorescnet level in the background .Conclusion Fluorescindiacetate could be used for staining cancer cell and detecting cell active substances such as active oxygens .
