Cavedilol%cardiomyopathy%Cx43%LVEF%adriamycin
目的:观察卡维地洛对阿霉素诱导的扩张型心肌病( dilated cardiomyopathy ,DCM)大鼠心室肌缝隙连接蛋白43(connexin 43,Cx43)表达的影响。方法:应用多柔比星腹腔注射建立DCM大鼠模型;模型组大鼠按照不同的给药方法随机分为卡维地洛组和模型对照组,卡维地洛组应用卡维地洛加双蒸馏水灌胃,模型对照组和正常对照组用定量的双蒸馏水灌胃,三组均1次/d,灌胃4周。12周末超声测量各组大鼠心脏左心室射血分数( LVEF),逆转录聚合酶链反应法检测Cx43 mRNA,采用免疫组化方法观察Cx43分布。结果:与模型对照组比较,卡维地洛组大鼠心肌Cx43 mRNA、Cx43平均灰度值、LVEF及均明显增高,卡维地洛组较模型对照组心肌Cx43的分布紊乱有改善,差异均有统计学意义。结论:卡维地洛能改善DCM大鼠心肌Cx43表达和分布,并能改善其心功能。

목적:관찰잡유지락대아매소유도적확장형심기병( dilated cardiomyopathy ,DCM)대서심실기봉극련접단백43(connexin 43,Cx43)표체적영향。방법:응용다유비성복강주사건립DCM대서모형;모형조대서안조불동적급약방법수궤분위잡유지락조화모형대조조,잡유지락조응용잡유지락가쌍증류수관위,모형대조조화정상대조조용정량적쌍증류수관위,삼조균1차/d,관위4주。12주말초성측량각조대서심장좌심실사혈분수( LVEF),역전록취합매련반응법검측Cx43 mRNA,채용면역조화방법관찰Cx43분포。결과:여모형대조조비교,잡유지락조대서심기Cx43 mRNA、Cx43평균회도치、LVEF급균명현증고,잡유지락조교모형대조조심기Cx43적분포문란유개선,차이균유통계학의의。결론:잡유지락능개선DCM대서심기Cx43표체화분포,병능개선기심공능。
Objective To observe the effect of Cavedilol on connexin 43 in dilated cardiomyopathy ( DCM) rats induced by adriamycin .Methods Intraperitoneal injection was used to prepare adriamycin induced dilated cardiomyopathy model in Wistar rats .The rats in the model group according to different methods of administration were randomly divided into Cavedilol group and the model control group .Their LVEF were investigated with echo-cardiogram.Immunohistochemical and image analytic techniques were used to observe myocardial Cx 43 expression and distribution in different group .The Cx43 mRNA level of cells was tested by reverse transcriptase polymerase chain reaction (RT-PCR) in different group.Results Compared with the model control group ,The Cx43 level of protein, the Cx43 mRNA level of cells and LVEF increased significantly in Cavedilol group ,the distribution of myo-cardial Cx43 could improve the disorder in Cavedilol group ,Comparing the differences among the 3 groups were sta-tistically significant .Conclusion Cavedilol can improve myocardial Cx 43 expression and distribution of DCM rats, and can improve cardiac function .
