目的:观察益气养肾方对糖尿病肾病大鼠应激反应水平的影响。方法选择10周龄SD大鼠50只,选取其中10只作为对照组,给予常规低脂、低糖饮食。其余40只采用高脂、高糖喂养联合低剂量(30mg/kg)链脲佐菌素诱导糖尿病肾病大鼠动物模型。建模成功后随机分为模型组与观察组各20只。观察组在模型组饮食基础上将药物益气养肾方按人与大鼠体表面积折算动物等效药量,按生药20g·kg-1·d-1,灌胃,连续灌胃3周。模型组同期继续给予高脂、高糖饮食。3周后杀死大鼠,手术摘取肾脏。病理切片观察3组肾脏组织形态学变化,比较3组大鼠肾脏肥大指数( kW/BW)、尿蛋白排泄量(24hUP)、血肌酐( Scr)水平、肾皮质丙二醛( MDA)含量、肾皮质超氧化物歧化酶(SOD)活性、肾皮质谷胱甘肽过氧化物酶(GSH-Px)。结果3组大鼠光镜下观察:对照组大鼠肾小球及肾小管正常,基底膜及系膜区结构清楚。模型组大鼠HE染色见肾小球肥大,肾小球毛细血管基底膜不规则增厚,肾小管上皮细胞肥大,内有糖原沉积表现,肾小管间纤维变性。观察组光镜观察可见与模型组相似病理改变,但肾小球肥大及肾小管肿胀较轻,蛋白管型较少。模型组及观察组kW/BW、24hUP、Scr较对照组明显升高,差异均有统计学意义(P<0.05),但观察组kW/BW、24hUP、Scr较模型组明显减少,差异均有统计学意义( P<0.05)。模型组及观察组MDA含量明显升高,SOD及GSH-Px较对照组明显降低,差异均有统计学意义( P<0.05)。观察组MDA含量较模型组明显减少,SOD、GSH-Px较模型组明显增加,差异均有统计学意义( P<0.05)。结论益气养肾方可能通过抑制糖尿病肾病大鼠体内氧化应激反应来减轻肾脏损伤,改善糖尿病肾病大鼠肾脏结构及起到功能保护作用。

목적:관찰익기양신방대당뇨병신병대서응격반응수평적영향。방법선택10주령SD대서50지,선취기중10지작위대조조,급여상규저지、저당음식。기여40지채용고지、고당위양연합저제량(30mg/kg)련뇨좌균소유도당뇨병신병대서동물모형。건모성공후수궤분위모형조여관찰조각20지。관찰조재모형조음식기출상장약물익기양신방안인여대서체표면적절산동물등효약량,안생약20g·kg-1·d-1,관위,련속관위3주。모형조동기계속급여고지、고당음식。3주후살사대서,수술적취신장。병리절편관찰3조신장조직형태학변화,비교3조대서신장비대지수( kW/BW)、뇨단백배설량(24hUP)、혈기항( Scr)수평、신피질병이철( MDA)함량、신피질초양화물기화매(SOD)활성、신피질곡광감태과양화물매(GSH-Px)。결과3조대서광경하관찰:대조조대서신소구급신소관정상,기저막급계막구결구청초。모형조대서HE염색견신소구비대,신소구모세혈관기저막불규칙증후,신소관상피세포비대,내유당원침적표현,신소관간섬유변성。관찰조광경관찰가견여모형조상사병리개변,단신소구비대급신소관종창교경,단백관형교소。모형조급관찰조kW/BW、24hUP、Scr교대조조명현승고,차이균유통계학의의(P<0.05),단관찰조kW/BW、24hUP、Scr교모형조명현감소,차이균유통계학의의( P<0.05)。모형조급관찰조MDA함량명현승고,SOD급GSH-Px교대조조명현강저,차이균유통계학의의( P<0.05)。관찰조MDA함량교모형조명현감소,SOD、GSH-Px교모형조명현증가,차이균유통계학의의( P<0.05)。결론익기양신방가능통과억제당뇨병신병대서체내양화응격반응래감경신장손상,개선당뇨병신병대서신장결구급기도공능보호작용。
Objective To observe the effect of yiqiyangshen recipe on the oxidative stress in diabetic nephropathy rats. Methods 50 SD rats with10-week old were selected,of which 10 rats were selected as the control group,and treated with conventional low-fat,low sugar diet. The remaining 40 rats with high fat and high glucose feeding(30mg/kg)combined with low dose of streptozotocin induced diabetic nephropathy rat model. After the success of modeling,40 rats were randomly divided into model group and observation group,each of 20. The observation group was treated with yiqiyangshen medcine. Model group was given high fat and high sugar diet. After 3 weeks,the rats were killed and the kidneys were removed,and then observed re-nal morphology changes and compared kW/BW,24hUP,Scr,MDA,SOD,GSH-Px level. Results The glomerulus and renal tubule of control group were all right,basement membrane and mesangial area structure clear. HE staining of the rats in the model group showed the glomerular hypertrophy,glomerular basement membrane thickness,renal tubular epithelial cell hyper-trophy,contained glycogen deposition performance,renal tubule fibrosis. Under light microscope,the pathological changes of model group and observation group were similar,but the glomerular hypertrophy and renal tubule swelling were lighter,and less protein tube. Compared with normal control group,kW/BW,24hUP and Scr of model group and observation group were signifi-cantly increased(P<0. 05),but compared with the model group,the kW/BW of observation group was significantly reduced, 24hUP and Scr of observation group was reduced significantly(P<0. 05). Compared with normal control group,renal cortical MDA content of model group and observation group increased obviously,SOD and GSH-Px decreased(P<0. 05). Compared with the model group,MDA content of observation group decreased significantly,SOD and GSH-Px increased significantly. Conclusion Yiqiyangshen recipe may reduce the renal injury by inhibiting oxidative stress reaction in diabetic nephropathy rats,and improve the renal structure in diabetic nephropathy rats and protect its function.
