目的:观察高胆红素模型新生豚鼠耳蜗核谷氨酸(glutamicacid,Glu)和γ-氨基丁酸(gamma-aminobutyric acid,GABA)含量的变化,探讨Glu和GABA在高胆红素损伤模型低位听觉中枢耳蜗核中的作用和意义。方法选择正常新生豚鼠30只随机分为三组:对照组、低胆红素(100μg/g)处理组和高胆红素(200μg/g)处理组,8h后处死动物取材处理标本免疫荧光观察各组耳蜗核Glu和GABA含量,Western blotting(线粒体凋亡相关蛋白检测)观察细胞质细胞色素C含量变化。结果与对照组相比,高浓度和低浓度胆红素处理组耳蜗核中GABA的含量水平明显下降,差异有显著性(p<0.05),而Glu的含量水平则明显上升(p<0.01);Western blotting显示细胞色素C在胆红素高低浓度处理组表达明显升高。结论新生豚鼠高胆红素模型Glu和GABA含量的动态变化反映了神经元的活动状况,提示二者在胆红素耳神经毒性中可能起重要作用。细胞质细胞色素C增高说明高胆红素可能介导神经细胞早期凋亡。

목적:관찰고담홍소모형신생돈서이와핵곡안산(glutamicacid,Glu)화γ-안기정산(gamma-aminobutyric acid,GABA)함량적변화,탐토Glu화GABA재고담홍소손상모형저위은각중추이와핵중적작용화의의。방법선택정상신생돈서30지수궤분위삼조:대조조、저담홍소(100μg/g)처리조화고담홍소(200μg/g)처리조,8h후처사동물취재처리표본면역형광관찰각조이와핵Glu화GABA함량,Western blotting(선립체조망상관단백검측)관찰세포질세포색소C함량변화。결과여대조조상비,고농도화저농도담홍소처리조이와핵중GABA적함량수평명현하강,차이유현저성(p<0.05),이Glu적함량수평칙명현상승(p<0.01);Western blotting현시세포색소C재담홍소고저농도처리조표체명현승고。결론신생돈서고담홍소모형Glu화GABA함량적동태변화반영료신경원적활동상황,제시이자재담홍소이신경독성중가능기중요작용。세포질세포색소C증고설명고담홍소가능개도신경세포조기조망。
Objective To study glutamate (glutamic acid, Glu) and gamma-aminobutyric acid (γ-GABA) expression levels in the cochlear nucleus of newborn guinea pigs with hyperbilirubinemia and effects of GABA and Glu on hearing dam?age. Methods Newborn guinea pigs were randomly divided into three groups:the control, and low and high bilirubin groups. Immunofluorescence was used to examine cochlear Glu and GABA levels;Western blotting analysis was used showing chang?es in cytochrome C expression. Results Compared with the control, GABA was significantly decreased (p<0.05) in both high and low bilirubin groups, whereas the levels of Glu were significantly increased (p<0.01). Western blotting showed significant?ly increased cytochrome C expression in bilirubin groups. Conclusion the dynamic changes of Glu and GABA in neonatal guinea pigs with hyperbilirubinemia reflects the status of neurons activities, suggesting that both may play important roles in bilirubin neurotoxicity affecting hearing. Cytochrome C increase in cytoplasm suggests that hyperbilirubinemia may induce early neurocyte aoptosis.
