目的:探讨B细胞淋巴瘤因子9(B-cell lymphoma 9, BCL9)在肝细胞肝癌(hepatocellular carcinoma, HCC)组织中的表达情况及临床意义。方法:采用Western Blot法检测8对新鲜冰冻HCC和对应癌旁组织中BCL9的蛋白表达;采用免疫组织化学法检测100例HCC和对应癌旁组织石蜡切片中BCL9的表达情况,并分析HCC中BCL9的表达与临床病理参数之间的关系。结果:Western Blot结果显示BCL9在HCC中的表达显著高于对应癌旁组织;免疫组化结果显示BCL9在HCC中主要定位于细胞核和细胞质,呈高表达,而在对应癌旁组织中主要定位于细胞质,表达较低或缺失;BCL9在HCC中的表达与肿瘤的分化程度和淋巴侵犯相关(P<0.05),与患者的性别、年龄、肿瘤直径、包膜完整、门静脉癌栓、血清HBsAg、甲胎蛋白及肝硬化无关(P>0.05)。结论:BCL9在HCC中高表达,可能参与了HCC的发生、发展及转移。
目的:探討B細胞淋巴瘤因子9(B-cell lymphoma 9, BCL9)在肝細胞肝癌(hepatocellular carcinoma, HCC)組織中的錶達情況及臨床意義。方法:採用Western Blot法檢測8對新鮮冰凍HCC和對應癌徬組織中BCL9的蛋白錶達;採用免疫組織化學法檢測100例HCC和對應癌徬組織石蠟切片中BCL9的錶達情況,併分析HCC中BCL9的錶達與臨床病理參數之間的關繫。結果:Western Blot結果顯示BCL9在HCC中的錶達顯著高于對應癌徬組織;免疫組化結果顯示BCL9在HCC中主要定位于細胞覈和細胞質,呈高錶達,而在對應癌徬組織中主要定位于細胞質,錶達較低或缺失;BCL9在HCC中的錶達與腫瘤的分化程度和淋巴侵犯相關(P<0.05),與患者的性彆、年齡、腫瘤直徑、包膜完整、門靜脈癌栓、血清HBsAg、甲胎蛋白及肝硬化無關(P>0.05)。結論:BCL9在HCC中高錶達,可能參與瞭HCC的髮生、髮展及轉移。
목적:탐토B세포림파류인자9(B-cell lymphoma 9, BCL9)재간세포간암(hepatocellular carcinoma, HCC)조직중적표체정황급림상의의。방법:채용Western Blot법검측8대신선빙동HCC화대응암방조직중BCL9적단백표체;채용면역조직화학법검측100례HCC화대응암방조직석사절편중BCL9적표체정황,병분석HCC중BCL9적표체여림상병리삼수지간적관계。결과:Western Blot결과현시BCL9재HCC중적표체현저고우대응암방조직;면역조화결과현시BCL9재HCC중주요정위우세포핵화세포질,정고표체,이재대응암방조직중주요정위우세포질,표체교저혹결실;BCL9재HCC중적표체여종류적분화정도화림파침범상관(P<0.05),여환자적성별、년령、종류직경、포막완정、문정맥암전、혈청HBsAg、갑태단백급간경화무관(P>0.05)。결론:BCL9재HCC중고표체,가능삼여료HCC적발생、발전급전이。
Objective:To investigate the expression and clinical significance of B-cell lymphoma 9(BCLP) in hepatocellular carcinoma(HCC). Methods:Western Blot was performed to detect the protein expression of BCL9 in 8 pairs of HCC tissues and adjacent nontumorous tissues. Immunohistochemistry technique was used to analyze the expression of BCL9 in 100 cases of HCC and adjacent nontumorous tissues. The relationship between BCL9 and clinical pathology parameters were analyzed statistically. Results:Western Blot results showed that the expression of BCL9 was observably higher in HCC than that in adjacent nontumorous tissues. Immunohistochemistry analysis showed that BCL9 in HCC mainly located in nucleus and cytoplasm, and it showed high expression there, while in the adjacent nontumorous tissue it was mainly located in the cytoplasm , where the expression was low or absent . The expression of BCL9 was significantly correlated with tumor differentiation grade and lymphatic invasion(P<0.05), but had no association with patients’ gender, age, tumor diameter, intact tumor capsules, portal vein tumor thrombus, serum HBsAg, alpha fetoprotein or cirrhosis of liver (P>0.05). Conclusion: BCL9 was overexpressed in HCC, and may involved in the occurrence, development and metastasis of HCC.
