目的:研究川芎嗪对阿霉素( ADM)诱导的大鼠心肌重塑的影响及是否通过改变TGF-β1的表达产生作用。方法:SD大鼠随机分为正常对照组( NC组,n=8;生理盐水2 mL/kg腹腔注射)、模型组( ADM组,n=9;ADM 2 mg/kg腹腔注射)、川芎嗪治疗组( Lig组,n=11;ADM 2 mg/kg+磷酸川芎嗪20 mg/kg腹腔注射)3组。测定各组SD大鼠心重指数(CWI)、左室心脏重量指数(LVWI),光镜下观察心肌病理学变化,免疫组化方法检测各组大鼠心肌组织中Ⅰ、Ⅲ型心肌胶原的表达,Western blot方法检测各组大鼠心肌组织中TGF-β1蛋白的表达。结果:3组大鼠间的心肌纤维化病理改变不同,CWI、LVWI、心肌组织中Ⅰ、Ⅲ型心肌胶原蛋白、Ⅰ/Ⅲ型胶原蛋白比值及TGF-β1蛋白的表达差异有统计学意义(F=5.971、5.556、51.740、57.281、28.934和61.263,P<0.05)。与ADM组相比, Lig组大鼠的心肌纤维化病理改变明显减轻,CWI、LVWI、心肌组织中Ⅰ、Ⅲ型心肌胶原蛋白、Ⅰ/Ⅲ型胶原蛋白比值及TGF-β1蛋白表达明显下降(P<0.05);Lig组大鼠的心肌组织中Ⅰ、Ⅲ型心肌胶原蛋白、Ⅰ/Ⅲ型胶原蛋白比值及TGF-β1蛋白的表达仍高于NC组( P<0.05)。结论:川芎嗪能改善阿霉素诱导的大鼠心肌重塑,其机制可能与影响TGF-β1的表达有关。

목적:연구천궁진대아매소( ADM)유도적대서심기중소적영향급시부통과개변TGF-β1적표체산생작용。방법:SD대서수궤분위정상대조조( NC조,n=8;생리염수2 mL/kg복강주사)、모형조( ADM조,n=9;ADM 2 mg/kg복강주사)、천궁진치료조( Lig조,n=11;ADM 2 mg/kg+린산천궁진20 mg/kg복강주사)3조。측정각조SD대서심중지수(CWI)、좌실심장중량지수(LVWI),광경하관찰심기병이학변화,면역조화방법검측각조대서심기조직중Ⅰ、Ⅲ형심기효원적표체,Western blot방법검측각조대서심기조직중TGF-β1단백적표체。결과:3조대서간적심기섬유화병리개변불동,CWI、LVWI、심기조직중Ⅰ、Ⅲ형심기효원단백、Ⅰ/Ⅲ형효원단백비치급TGF-β1단백적표체차이유통계학의의(F=5.971、5.556、51.740、57.281、28.934화61.263,P<0.05)。여ADM조상비, Lig조대서적심기섬유화병리개변명현감경,CWI、LVWI、심기조직중Ⅰ、Ⅲ형심기효원단백、Ⅰ/Ⅲ형효원단백비치급TGF-β1단백표체명현하강(P<0.05);Lig조대서적심기조직중Ⅰ、Ⅲ형심기효원단백、Ⅰ/Ⅲ형효원단백비치급TGF-β1단백적표체잉고우NC조( P<0.05)。결론:천궁진능개선아매소유도적대서심기중소,기궤제가능여영향TGF-β1적표체유관。
Aim:To investigate whether change in TGF-β1 expression is involved in the mechanism by which ligus-trazine affects adriamycin-induced myocardial remodelling in rats .Methods: SD rats were randomly divided into normal control group(NC group,n=8), model group(ADM group,n=9), and ligustrazine treatment group (Lig group,n=11). For each group, cardiac weight index (CWI) and left ventricular weight index (LVWI) were measured, pathological chan-ges were observed by light microscopy , typeⅠ,Ⅲmyocardial collagen expression was determined by immunohistochemis-try, and TGF-β1 expression in cardiac tissue was detected by Western blot .Results: Myocardial fibrosis, CWI, LVWI, typeⅠ,Ⅲmyocardial collagen expression , ratio of Ⅰ/Ⅲ myocardial collagens and TGF-β1 protein expression showed significant differences among the three groups (F=5.971, 5.556, 51.740,57.281,28.934,61.263;P<0.05).Com-pared with ADM group, cardiac fibrosis was significantly alleviated , CWI, LVWI, typeⅠ,Ⅲmyocardial collagen expres-sion, ratio of type Ⅰ/Ⅲmyocardial collagen and TGF-β1 protein expression significantly decreased in the Lig group ( P<0.05);however, type Ⅰ,Ⅲmyocardial collagen expression , ratio of type Ⅰ/Ⅲ myocardial collagen, and TGF-β1 ex-pression in cardiac tissue of the Lig group were still higher than those of NC group (P<0.05).Conclusion:Ligustrazine can improve cardiac remodeling induced by adriamycin , and changes in TGF-β1 may be part of the mechanism .
